    MicroRNAs : An Emerging Player In Autophagy

    Autophagy is an evolutionarily conserved self-digestion process for the quality control of intracellular entities in eukaryotes. In the past few years, mounting evidence indicates that microRNAs (miRNAs)-mediated post-transcriptional regulation of gene expression represents an integral part of the autophagy regulatory network and may have a substantial effect on autophagy-related physiological and pathological conditions including cancer. Herein, we examine some of the molecular mechanisms by which microRNAs manipulate the autophagic machinery to maintain cellular homeostasis and their biological outputs during cancer development. A better understanding of interaction between microRNAs and cellular autophagy may ultimately benefit future cancer diagnostic and anticancer therapeutics.

      Yang,et.al discussed the microRNA regulation of autophagy briefly in light of the current literature. They elegantly discussed the role of microRNAs according to the stages of autophagy and their role in chemotherapy. The text is fluent and discussing the facts well enough for this kind of short review.
      Although the text is well written, the figure poorly represents the information presented in the text and there is a need for seperate table.
      Therefore, the manuscript can be revised according to the following comments:

      Major comments:
      1- Autophagic machinery is very superficially explained in this figure. It will better to have a more detailed version without the miRNAs involved.
      -mTOR pathway can be briefly presented in the figure.
      -p62 is missing in the figure.
      -ATG9 is shown not on the figure but only in the box.
      -Other RAB proteins can be mentioned in the figure also. (RAB5 is present in text but not in figure)
      2-miRNAs should be presented in a seperate table, where their targeting should be shown as direct or indirect (direct- luciferase)
      3-There are missing miRNAs in the text, due to the publication date of the article. Since December more than 20 new articles have been published. Authors may include this literature to their text.
      4- There are inconsistencies between the text and figure. Also, some works were not cited in the text (Literature before Dec 2014). Authors can update their list in a comprehensive way.
      5-In conclusion, authors mentioned role of autophagy in microRNA biogenesis by regulating AGO and DICER. The discussion could be more detailed.

      Minor comments:
      1- Please check for typing mistakes.
      pg.1 mTOC for instance
      2- Separate the figure and the table or integrate ATG9 for example in the figure
