          Abstract

          Abstract: The digital image analysis of seedlings has become largely employed in seed quality-control programs due to its feasibility, objectivity and fast results. Despite these advantages, no studies have yet demonstrated the efficiency of this technique for analyzing coffee seed vigor. The present study aimed at evaluating the efficiency of the Seed Vigor Imaging System (SVIS®) in determining the vigor of coffee seeds (Coffea arabica L.) from digital images of seedlings. Also, the results of these analyses were contrasted with conventional vigor tests. Six seed lots from each cultivar, Bourbon and Catucaí 20/15, were used. The research was conducted in two experimental times, and the seed vigor was determined by the tests of first germination count, accelerated aging, electrical conductivity, primary root protrusion speed, as well as by the analysis of scanned images of seedlings, made possible by the SVIS® software. For the Bourbon and Catucaí 20/15 cultivars, the SVIS® analysis was able to discriminate lots of both high and low vigor (vigor indexes of 317 to 752, and 181 to 703, respectively). A similar outcome was obtained from the conventional tests appraised in this research. Eventually, it was possible to conclude that the digital analysis of 20-day-old seedlings by SVIS® is an efficient method to evaluate coffee seed vigor.

          Resumo: A análise de imagens digitalizadas de plântulas é cada vez mais utilizada em programas de controle de qualidade de sementes devido à sua praticidade, objetividade e rapidez na obtenção dos resultados. Apesar dessas vantagens, ainda não existem pesquisas comprovando a eficiência dessa técnica para a análise do vigor de sementes de café. Assim, esta pesquisa teve como objetivo avaliar a eficiência da análise de imagens digitalizadas de plântulas pelo Seed Vigor Imaging System (SVIS®) na determinação do vigor de sementes de café (Coffea arabica L.), e compará-la com os testes convencionalmente empregados. Foram utilizadas sementes das cultivares Bourbon e Catucaí 20/15, cada cultivar representada por seis lotes. A pesquisa foi conduzida em duas épocas de avaliação e o vigor determinado por meio das avaliações de primeira contagem de germinação, envelhecimento acelerado, condutividade elétrica, velocidade de protrusão da raiz primária e análise de imagens digitalizadas de plântulas com o SVIS®. Tanto para a cultivar Bourbon como para a cultivar Catucaí 20/15, a análise SVIS® permitiu discriminar lotes com vigor mais baixo e mais alto (índices de vigor de 317 a 752, e 181 a 703, respectivamente), de maneira semelhante aos testes convencionais utilizados nesta pesquisa. Concluiu-se que a análise de imagens digitalizadas de plântulas com 20 dias de idade após a semeadura pelo SVIS® é eficiente na avaliação do vigor de lotes de sementes de café.

          Seed vigor testing: an overview of the past, present and future perspective

          The assessment of seed vigor has many important implications to the seed industry as a basic monitoring of seed physiological potential during different phases of seed production and a support for strategic decisions regarding the selection of high quality seedlots to meet the consumer demand. The potential attributes of seed vigor as a fundamental physiological seed characteristic and its association with field stand establishment and crop productivity has been worldwide recognized from the 1960s onward. This led to the diversification of research approaches involving the synchronization of different physiological characteristics and events that determine the potential for high performance during seed storage and after sowing. The basic objective of vigor testing is to provide a consistent identification of differences in physiological potential among seedlots of commercial value and this represents a more sensitive parameter than the germination test. There are various procedures to assess seed vigor including those that directly or indirectly evaluate seed metabolic state or identify seed tolerance to specific stress(es). At the same time, in more recent years, new knowledge has come from molecular biology, biotechnology, biophysics and seed and seedling imaging analyses; these approaches has been important complements to traditional seed research.. This text has the purpose to emphasize the role of Brazilian research in seed vigor, whose competence is internationally recognized, and to provide an overview of the evolution of knowledge also attempting to highlight events that contributed to the advancement of research on the subject.

            Métodos para avaliação do vigor de sementes de soja, incluindo a análise computadorizada de imagens

            A velocidade e uniformidade de emergência de plântulas, determinantes do sucesso do estabelecimento do estande, representam etapas essenciais para a obtenção de alta produtividade na cultura da soja, de modo que o desenvolvimento de procedimentos para avaliar o potencial fisiológico tem recebido atenção contínua da pesquisa em Tecnologia de Sementes. O objetivo deste trabalho foi o de verificar a eficiência de um sistema computadorizado para análise de imagens de plântulas, em comparação com testes recomendados para determinação do vigor de sementes de soja como testes de tetrazólio, envelhecimento acelerado, condutividade elétrica e crescimento de plântulas,. Para tanto, lotes de sementes dos cultivares Pintado e TMG 113RR foram avaliados quanto à germinação (25oC aos 4 e 7 dias), envelhecimento acelerado tradicional (41oC durante 48 h), envelhecimento acelerado com solução saturada de sal (41oC durante 72 h) e emergência de plântulas. O sistema de análise computadorizada de imagens de plântulas (SVIS®), desenvolvido pela Ohio State University/EUA, forneceu dados referentes ao índice de vigor, à uniformidade de desenvolvimento e ao crescimento de plântulas, tanto aos três dias após a semeadura do teste de germinação sob condições ótimas como após o envelhecimento acelerado com solução de sal. O período experimental compreendeu três épocas de testes com intervalo aproximado de 30 dias. Constatou-se que o SVIS® é suficientemente sensível para avaliação do vigor de sementes de soja, com eficiência comparável à do teste de envelhecimento acelerado, constituindo alternativa interessante para a obtenção de informações consistentes sobre o potencial fisiológico.

              Vigor-S, a new system for evaluating the physiological potential of maize seeds

              ABSTRACT: The refinement of vigor tests and the possibility of utilizing computer resources for the effective evaluation of the seed physiological potential have attracted considerable interest from research and seed technologists. The aim of this study was to evaluate the physiological potential of maize seeds using the newly-created Automated Analysis of Seed Vigor System (Vigor-S) compared with other recommended seed vigor tests; two maize hybrids were used, each represented by seven seed lots. Germination and vigor (cold test, saturated salt accelerated aging, and field seedling emergence) evaluations were conducted. For the evaluation of seed vigor with the use of seedling image analysis resources, two systems were compared: the Seed Vigor Imaging System (SVIS®), developed by Ohio State University, USA and the Vigor-S, resulting from collaboration between USP/ESALQ and EMBRAPA (Embrapa Instrumentation). Using these two systems, three day old seedlings were scanned and the images were analyzed. Similar results for the vigor index, uniformity of development, and seedling length were obtained. The computerized image analysis of seedlings using Vigor-S has advantages with respect to accuracy, speed, and the possibility of automatic application to a worksheet. It is a consistent alternative for the evaluation of maize seed vigor, and produces information compatible with that obtained by the accelerated aging test and SVIS®.
