          Abstract

          Trypanosoma vivax infecta uma grande variedade de animais ungulados selvagens e domésticos, podendo causar grande impacto na produção de ruminantes. Este trabalho teve como objetivo avaliar a detecção de anticorpos IgG anti-Trypanosoma vivax em bovinos provenientes do estado de Pernambuco, Brasil. Para tanto, foram analisadas 2,053 amostras de soro sanguíneo de bovinos provenientes de rebanhos de municípios do estado de Pernambuco, os quais foram analisados através da Reação de Imunofluorescência Indireta. Das amostras testadas 13,93% (286/2.053) foram reagentes para anticorpos IgG anti-Trypanosoma vivax. As freqüências, por mesorregião, variaram de 11,90% a 15,99%. Assim, os dados obtidos permitiram a caracterização do estado de Pernambuco como uma área de instabilidade enzoótica e sugere que o estado Pernambuco é área endêmica para Trypanosoma vivax e este parasito está distribuído por todo o estado.

          Translated abstract

          Trypanosoma vivax infects a wide range of wild and domestic ungulates, causing important losses for the livestock industry. The aim of the present study was to assess the detection of IgG antibodies against T. vivax in cattle from the state of Pernambuco, Brazil. Therefore, we analyzed 2.053 blood serum samples from cattle herds of municipalities in Pernambuco, what was made by Immunofluorescence Assay. The overall seroprevalence of IgG antibodies against T. vivax in cattle was 13.93% (286/2053). The frequencies, by region, varied from 11.90% to 15.99%. Thus, the data obtained allowed to characterize the state of Pernambuco as an area of enzootic instability for T. vivax. The frequency herein reported (i.e., 13.93%) indicates that Pernambuco is an endemic area for T. vivax, this parasite being spread throughout the state.

            Trypanosoma vivax: characterization of the spliced-leader gene of a Brazilian stock and species-specific detection by PCR amplification of an intergenic spacer sequence.

            The sequence of the spliced-leader gene repeat of a Brazilian Trypanosoma vivax stock from cattle showed high similarity to sequences of West African T. vivax in both intron and intergenic sequences. This is the first evidence based on DNA sequences of close-relatedness between Brazilian and West African T. vivax stocks. A T. vivax-specific diagnostic PCR assay based on spliced-leader gene intergenic sequences was able to amplify DNA from T. vivax stocks from South America (Brazil, Bolivia, and Colombia) and West Africa. Species-specificity of this method was confirmed by results obtained by testing 15 other trypanosomes, including other species and subspecies that can also infect cattle. The PCR assay developed presented high sensitivity, detecting the DNA content of only one parasite and also revealing T. vivax infection in asymptomatic animals without detectable parasitemia by microhematocrit or in Giemsa-stained blood smears. Use of crude preparations from field-blood samples collected on both filter paper and glass slides as DNA template suggested that this method could be useful for the diagnosis of T. vivax in large epidemiological studies.               First report and molecular characterization of Trypanosoma vivax in cattle from state of Pernambuco, Brazil.

              This paper describes an outbreak of Trypanosoma vivax for the first time in the state of Pernambuco, Brazil, affecting dairy cattle in the municipality of Itambé in the northern coastal zone of the state. Clinical signs compatible with infection by blood protozoa and epidemic miscarriages were observed. The diagnosis of T. vivax was confirmed through biometric microscopy and molecular analysis with PCR and DNA sequencing. The T. vivax isolate detected in the present study proved to be genetically very close to other Brazilian isolates of the protozoan despite being geographically distant.
