          Abstract

          We demonstrate maintenance and transmission of severe fever with thrombocytopenia syndrome virus by Haemaphysalis longicornis ticks in the larva, nymph, and adult stages with dissemination in salivary gland, midgut, and ovarian tissues. The H. longicornis tick is a competent vector to transmit this virus in both transovarial and transstadial modes.

          Biology, ecology and distribution of the tick, Haemaphysalis longicornis Neumann (Acari: Ixodidae) in New Zealand.

          Haemaphysalis longicornis is the only tick in New Zealand that infests livestock. Throughout its range H. longicornis is exposed to and exhibits tolerance to a wide range of environmental conditions, although it flourishes more in moist, warm-temperate environments. This review examines aspects of the biology, physiology and ecology of H. longicornis that determine its distribution and seasonal activity in New Zealand, based on laboratory and field studies. Examples are also drawn from studies outside New Zealand for comparative purposes, especially in the context of seasonal activity as seen in less temperate latitudes. The tick is able to withstand a wide range of temperature, from its developmental threshold of ∼12°C to nearly 40°C at its lethal limit, but its tolerance of dehydration is less wide, especially in the larva and adult, the former especially being the stage that largely determines suitable biotopes for the tick and its present distributional limits. The importance of H. longicornis to the New Zealand livestock industry has recently increased through the establishment and spread of Theileria orientalis Ikeda among dairy and beef cattle, although the tick has always posed production-limiting problems for cattle, deer and to a lesser extent, sheep. The tick's role as a vector of theileriosis and how aspects of the tick's biology affect the spread and maintenance of this disease are discussed. It is proposed that, of available wildlife hosts, the brown hare with its wide-ranging habits, is an important disseminator of ticks. Currently control of ticks is difficult partly because of their wide host range, overlapping activity periods of stadia, and also because the greater part of their annual cycle is spent on pasture. This means that acaricides alone do not satisfactorily reduce tick populations or provide comprehensive protection to stock, so integrated management combining pasture management with good husbandry and chemical prophylaxis is advocated.

            Prevalence of severe fever with thrombocytopenia syndrome virus in Haemaphysalis longicornis ticks in South Korea.

            Haemaphysalis longicornis a vector that harbors severe fever with thrombocytopenia syndrome virus (SFTSV) is a major species of tick in South Korea. To investigate the existence and prevalence of SFTSV in Korea, we collected ticks from nine provinces in South Korea for detecting SFTSV. In all, we collected 13,053 ticks, and H. longicornis (90.8%, 11,856/13,053) was the most abundant among them. The minimum infection rate (MIR) of SFTSV in H. longicornis was 0.46% (55 pools). SFTSV was detected in ticks during all the developmental stages, showing MIR in larvae (2/350, 0.57%), nymphs (38/10,436, 0.36%), males (2/221, 0.90%), and females (13/849, 1.53%), respectively. Viruses were detected in ticks collected between April and September. A higher MIR was detected in ticks from the southern part of the country. We amplified the M and S segment partial genes from a sample and analyzed the nucleotide sequence. The results showed a 93-98% homology to Chinese and Japanese strains registered in Genbank. In this study, we confirmed the existence of SFTSV for the first time in South Korea. The SFTSV prevalence data from the studies are essential for raising the awareness of SFTS in South Korea.

              Detection of Theileria orientalis genotypes in Haemaphysalis longicornis ticks from southern Australia

              Background Theileria are blood-borne intracellular protozoal parasites belonging to the phylum Apicomplexa. Previously considered a benign parasite in Australia, outbreaks of clinical disease resulting from Theileria orientalis genotypes have been reported in Australia since 2006. Since this time, outbreaks have become widespread in south-eastern Australia, resulting in significant adverse impacts on local dairy and beef industries. This paper provides the first investigation into the possible biological and mechanical vectors involved in the rapid spread of the parasite. Methods To identify possible vectors for disease, ticks, biting flies and mosquitoes were collected within active outbreak regions of Gippsland, Victoria. Ticks were collected from cattle and wildlife, and mosquitoes and biting flies were collected in traps in close proximity to outbreak herds. Ticks were identified via DNA barcoding of the mitochondrial cytochrome oxidase I gene. Barcoded ticks were pooled according to species or phylogenetic group and tested for the presence of T. orientalis and the genotypes Ikeda, Chitose and Buffeli using real-time PCR. Results DNA barcoding and phylogenetic analysis identified ticks from the following species: Haemaphysalis longicornis, Ixodes holocyclus, Ixodes cornuatus, Ixodes hirsti, and Bothriocroton concolor. Additional Haemaphysalis, Ixodes and Bothriocroton spp. were also identified. Of the ticks investigated, only H. longicornis ticks from cattle carried theilerial DNA, with the genotypes Ikeda, Chitose and Buffeli represented. Mosquitoes collected in close proximity to outbreak herds included; Aedes camptorhynchus, Aedes notoscriptus, Coquillettidia linealis, Culex australicus, and Culex molestus. Low levels of T. orientalis Buffeli genotype were detected in some mosquitoes. The haematophagous flies tested negative. Conclusions This is the first demonstration of a potential vector for T. orientalis in the current Australasian disease outbreak.
