      Environmental Effects on Compulsive Tail Chasing in Dogs

          Abstract

          Obsessive Compulsive Disorder (OCD) is a neuropsychiatric disorder observed both in humans and animals. Examples of Canine Compulsive Disorder (CD) include excessive tail chasing (TC), light/shadow chasing and flank sucking. We performed a questionnaire survey to investigate the characteristics of compulsive (TC) and its possible associations with environmental correlates and personality in a pet population of 368 dogs from four dog breeds. We observed an early onset of TC at 3–6 months of age and a large variation in TC frequency in all breeds, with an overrepresentation of milder cases. Almost half of the TC dogs showed lowered responsiveness during bouts and displayed also other types of compulsions more often than the controls. Interestingly, dogs that received dietary supplements, especially vitamins and minerals, expressed less TC compared to dogs that did not receive any supplements. Neutered females had less TC, suggesting an influence of ovarian hormones on TC. Tail chasers were shyer and had separated earlier from their mothers than the controls. Finally, our genetic study did not find an association between TC and CDH2, a locus previously associated with the canine flank sucking compulsion. In conclusion, the early-onset and the variable nature of the repetitive behaviour, which is affected by environmental factors such as micronutrients, neutering and maternal care, share several similar components between canine and human compulsions and supports canine TC as a model for human OCD.

          Common genetic variants on 5p14.1 associate with autism spectrum disorders.

          Autism spectrum disorders (ASDs) represent a group of childhood neurodevelopmental and neuropsychiatric disorders characterized by deficits in verbal communication, impairment of social interaction, and restricted and repetitive patterns of interests and behaviour. To identify common genetic risk factors underlying ASDs, here we present the results of genome-wide association studies on a cohort of 780 families (3,101 subjects) with affected children, and a second cohort of 1,204 affected subjects and 6,491 control subjects, all of whom were of European ancestry. Six single nucleotide polymorphisms between cadherin 10 (CDH10) and cadherin 9 (CDH9)-two genes encoding neuronal cell-adhesion molecules-revealed strong association signals, with the most significant SNP being rs4307059 (P = 3.4 x 10(-8), odds ratio = 1.19). These signals were replicated in two independent cohorts, with combined P values ranging from 7.4 x 10(-8) to 2.1 x 10(-10). Our results implicate neuronal cell-adhesion molecules in the pathogenesis of ASDs, and represent, to our knowledge, the first demonstration of genome-wide significant association of common variants with susceptibility to ASDs.

            The sleep disorder canine narcolepsy is caused by a mutation in the hypocretin (orexin) receptor 2 gene.

            Narcolepsy is a disabling sleep disorder affecting humans and animals. It is characterized by daytime sleepiness, cataplexy, and striking transitions from wakefulness into rapid eye movement (REM) sleep. In this study, we used positional cloning to identify an autosomal recessive mutation responsible for this sleep disorder in a well-established canine model. We have determined that canine narcolepsy is caused by disruption of the hypocretin (orexin) receptor 2 gene (Hcrtr2). This result identifies hypocretins as major sleep-modulating neurotransmitters and opens novel potential therapeutic approaches for narcoleptic patients.

              Development and validation of a questionnaire for measuring behavior and temperament traits in pet dogs.

              To develop and validate a questionnaire to assess behavior and temperament traits of pet dogs. Cross-sectional survey of dog owners. Animals-1,851 dogs belonging to clients of a veterinary teaching hospital or members of national breed clubs and 203 dogs examined by canine behavior practitioners because of behavior problems. Owners were asked to complete a questionnaire consisting of 152 items eliciting information on how dogs responded to specific events and situations in their usual environment. Data from completed questionnaires were subjected to factor analysis, and the resulting factors were tested for reliability and validity. Factor analysis yielded 11 factors from 68 of the original questionnaire items that together accounted for 57% of the common variance in questionnaire item scores. Reliability was acceptable for all but 1 of these factors. Behavior problems in 200 of the 203 dogs with behavior problems could be assigned to 7 diagnostic categories that matched 7 of the factors identified during factor analysis of questionnaire responses. Dogs assigned to particular diagnostic categories had significantly higher scores for corresponding questionnaire factors than did those assigned to unrelated diagnostic categories, indicating that the factors were valid. Validity of the remaining 4 factors could not be examined because of a lack of information on dogs with behavior problems related to these factors. Findings suggest that the resulting 68-item questionnaire is a reliable and valid method of assessing behavior and temperament traits in dogs. The questionnaire may be useful in screening dogs for behavior problems and in evaluating the clinical effects of various treatments for behavior problems.
