      Serotypes and molecular typing analysis of human Salmonella in Zhangjiagang, Jiangsu, 2017

          Abstract

          Objective To analyze the serotypes characteristics of human Salmonella in Zhangjiagang in 2017 through molecular typing of dominant serogroups determined by pulsed-field gel electrophoresis (PFGE), and accumulate the data for Salmonella type libraries in the region.

          Methods In 2017, the serological characteristics of human Salmonella in Zhangjiagang area were collected and extracted, PFGE molecular typing was carried out for the dominant group B Salmonella, and the correlation fingerprint was obtained and clustered by PulseNet China network.

          Results 92 strains of Salmonella isolates belong to 16 serotypes in 7 serogroups, dominated with serogroup B (33.70%), and 30 strains of serogroup B Salmonella PFGE fingerprints can be divided into 26 genotypes (similar values 100% for the same PFGE type) and 19 clones (clone lines of the same PFGE with similarity between 0.85-1.00). Among them, 9 strains of S. derby can be divided into 5 clones and 7 genotypes with the similarity of 60.92%-100%; 7 strains of PFGE fingerprints of S.agona were 5 clones and 6 genotypes with the similarity of 0 -100%; 6 strains of S. typhimurium can be divided into 3 clones and 5 genotypes, with the similarity of 48.51%一 100%.

          Conclusion The serotypes of human Salmonella in Zhangjiagang are diverse, and the Salmonella serogroup Bis the dominant serogroup. The results of PFGE molecular typing showed there were many PFGE bands and high gene polymorphism in Salmonella. Different PFGE fingerprints could be obtained from the same serotype. Among them, the phylogenetic relationship between S. derby was close, and some strains of S. typhimurium were closely related. There were significant differences in the clonal lines of S.agona, and the genetic relationship was far.

          摘要: 目的 分析 2017 年张家港检出的 92 株人源沙门菌的血清学特征, 用脉冲场凝胶电泳 (PFGE) 技术对优势血 清群进行分子分型研究, 为该地区沙门菌菌型库积累数据。 方法 收集和提取 2017 年张家港地区人源沙门菌患者血 清学特征, 对优势群 B 群沙门菌进行PFGE分子分型检测, 利用 PulseNet China 网络获得相关性指纹图谱并进行聚类分 析。 结果 92 株沙门菌分离株分布 7 个血清群和 16 个血清型, 以 B 群为主 (33.70%), 30 株 B 群沙门菌 PFGE 指纹图谱 可分为 26 个基因型(相似值 100% 为同一 PFGE 型别) , 19 个克隆系 (相似度在 0.85~1.00 的为同一 PFGE 的克隆系) 。其 中 9 株德尔卑沙门菌可分为 5 个克隆系 7 个基因型, 7 个基因型相似度 60.92%~100%; 7 株阿贡那沙门菌 PFGE 指纹图 谱 5 个克隆系 6 个基因型, 6 个基因型相似度 0~100%;6 株鼠伤寒沙门菌可分为 3 个克隆系 5 个基因型, 5 个基因型相似 度 48.51%~100%。 结论 张家港市人源沙门菌血清型多样化分布, B 群沙门菌为优势血清群。沙门菌 PFGE 带型较 多, 基因多态性较高, 同一血清型可获得不同 PFGE 指纹图谱。其中德尔卑沙门菌亲缘关系近, 鼠伤寒部分菌株存在较 近的亲缘关系, 阿贡那沙门菌各克隆系存在明显差异, 亲缘关系较远。
