          Abstract

          RESUMEN El objetivo del estudio fue caracterizar el semen del Perro sin Pelo del Perú. Quince machos, de edades entre 1 a 5 años, inscritos en el Kennel Club Peruano fueron utilizados para colectar semen (un eyaculado por macho) mediante manipulación digital. Se realizó un espermatograma básico y se registraron características macroscópicas (volumen y color) y microscópicas (motilidad y concentración espermática, utilizando microscopía de campo claro). Adicionalmente, se realizaron pruebas de función espermática: viabilidad espermática (utilizando SYBR-14 y ioduro de propidio), potencial de membrana mitocondrial (PMM; utilizando MitoTraker Deep Red FM), e integridad acrosomal (utilizando FITC-PSA y ioduro de propidio), analizadas posteriormente mediante citometría de flujo con análisis de imágenes. Se encontraron los siguientes valores (medias ± IC 95%): 6.1 ± 2.7 ml de volumen, color blanquecino; 66.7 ± 10.5% de motilidad; 262.7 ± 20.4 x 106 espermatozoides/ml de concentración espermática; 77.7 ± 9.7% de viabilidad espermática; 85.9 ± 5.1% de alto PMM; 83.6 ± 14.9% de integridad acrosomal. Estos resultados representan el primer reporte sobre las características seminales de esta raza y podrían ser tomados como valores de referencia para una mejor selección de machos reproductores y para el diagnóstico de alteraciones reproductivas de la raza.

          Translated abstract

          ABSTRACT The aim of this study was to characterize the semen of the Peruvian Hairless Dog. Fifteen males, aged 1 to 5 years, registered by the Kennel Club Peruano were used to collect semen (one ejaculated per male) by digital manipulation. A basic spermatogram was performed and macroscopic (volume and colour) and microscopic (sperm motility and concentration, using light field microscopy) characteristics were recorded. Additionally, sperm function tests were performed: sperm viability (using SYBR-14 and propidium iodide), mitochondrial membrane potential (MMP, using MitoTraker Deep Red FM), and acrosomal integrity (using FITC-PSA and propidium iodide) and analysed by flow cytometry with image analysis. The following values were found (mean ± 95% CI): 6.1 ± 2.7 ml volume, opalescent colour; 66.7 ± 10.5% motility; 262.7 ± 20.4 x 106 sperm/ml sperm concentration; 77.7 ± 9.7% of sperm viability; 85.9 ± 5.1% high MMP; 83.6 ± 14.9% acrosomal integrity. These results represent the first report on the seminal characteristics of this breed and could be taken as reference values for a better selection of breeding males and for the diagnosis of reproductive alterations of the breed.

          Semen collection in the dog.

          This review will discuss semen collection in the dog. Semen samples may be collected from male dogs for the purposes of artificial insemination, cryopreservation or diagnosis. The materials needed for semen collection depend on which method is used and the collector's level of expertise with this procedure. At minimum, two sterile centrifuge tubes or specimen cups can be used to collect semen as it is ejaculated (for the combined first and second fractions and for the third fraction). The most common method for semen collection in the dog is by digital stimulation. Under ideal conditions, this procedure is performed in the presence of an estrous bitch. Initially, the dog's penis is vigorously massaged through the prepuce at the level of the bulbus glandis (caudal-most aspect of the prepuce) until a partial erection develops (initial engorgement of the bulbus glandis). The prepuce is quickly retracted past the bulbus glandis and firm constant pressure is applied to the penis behind the bulbus glandis by squeezing the penis between index finger and thumb. Pelvic thrusting may occur following application of pressure behind the bulbus glandis during the development a "full" erection. The ejaculate is composed of three fractions: first (sperm-poor), second (sperm-rich) and third (prostatic fluid). In addition to digital stimulation of the penis, spermatozoa have been collected from dogs using electroejaculation and pharmacologic methods.

            The value of canine semen evaluation for practitioners.

            Compared to human medicine, little is known in canine medicine regarding specific findings on semen evaluation and their correlation with fertility. Suggestions to optimize quality of semen evaluation in veterinary practice include creating standardized protocols for evaluation of all semen parameters and updating those protocols as needed; creating some form of quality control for the clinic laboratory; educating owners about our inability to predict with 100% accuracy whether dogs with poor semen quality never could impregnate a bitch or whether dogs with excellent semen quality always could impregnate a bitch; and generating protocols for further diagnostic work-up for those dogs with abnormal semen quality.
