          Abstract

          Resumen El diclofenaco sódico y el ibuprofeno son medicamentos de alto consumo, empleados para el alivio de dolencias comunes y su poca metabolización hace que gran proporción de estos se conviertan en contaminantes acuíferos, que por su bioacumulación representan una problemática ambiental que requiere atención urgente. Es por esto que el seguimiento de estos contaminantes, a través del desarrollo y validación de metodologías analíticas que permitan su cuantificación en fuentes hídricas, se convierten en una necesidad para su prevención y control. En este estudio se desarrolló un método por cromatografía líquida de alta resolución para la determinación simultánea de diclofenaco e ibuprofeno en aguas. La validación del método se llevó a cabo con la determinación de la linealidad, límites de cuantificación y detección, exactitud y precisión en términos de repetibilidad y precisión intermedia. Para la determinación de los parámetros de desempeño se calculó el porcentaje de recuperación, el promedio, la desviación estándar, la desviación estándar relativa, el coeficiente de variación y el intervalo de confianza. Además, para determinar la exactitud y la precisión se aplicó el análisis factorial de varianza con un nivel de confianza del 95 % y nivel de significancia del 0,05. Los valores obtenidos para todos los parámetros cumplieron con los criterios de aceptación establecidos, con lo que se concluye que el método desarrollado es exacto y preciso con altos niveles de confiabilidad.

          Translated abstract

          Abstract Diclofenac sodium and ibuprofen are highly consumed drugs, used for the relief of common ailments, their poor metabolization means that a large proportion of them become aquifer contaminants, this problem requires urgent attention. The monitoring of these contaminants, through the development and validation of analytical methodologies that allow their quantification in water resources, becomes a necessity to prevent and control them. In this study, a high-performance liquid chromatography method was developed for the simultaneous determination of diclofenac and ibuprofen in water. The validation of the method was carried out with the determination of linearity, limits of quantification and detection, accuracy and precision in terms of repeatability and intermediate precision. For the determination of the performance parameters, the percentage recovery, average, standard deviation, relative standard deviation, coefficient of variation and confidence interval were calculated. In addition, to determine the accuracy and precision, the factorial analysis of variance was applied with a confidence level of 95 % and a significance level of 0.05. The values obtained for all parameters complied with established acceptance criteria, confirming that the method developed is accurate and precise with high levels of reliability.

O monitoramento destes contaminantes, através do desenvolvimento e validação de metodologias analíticas que permitem sua quantificação nos recursos hídricos, torna-se uma necessidade para preveni-los e controlá-los. Neste estudo, foi desenvolvido um método de cromatografia líquida de alto desempenho para a determinação simultânea de diclofenaco e ibuprofeno na água. A validação do método foi realizada com a determinação da linearidade, limites de quantificação e detecção, precisão e precisão em termos de repetibilidade e precisão intermediária. Para a determinação dos parâmetros de desempenho, foram calculados a porcentagem de recuperação, média, desvio padrão, desvio padrão relativo, coeficiente de variação e intervalo de confiança. Além disso, para determinar a precisão e a precisão, a análise fatorial de variância foi aplicada com um nível de confiança de 95 % e um nível de significância de 0,05. Os valores obtidos para todos os parâmetros cumpriram os critérios de aceitação estabelecidos, confirmando que o método desenvolvido é preciso e preciso com altos níveis de confiabilidade.

              Diclofenac and its transformation products: Environmental occurrence and toxicity - A review.

              Diclofenac (DCF) is a prevalent anti-inflammatory drug used throughout the world. Intensive researches carried out in the past few decades have confirmed the global ubiquity of DCF in various environmental compartments. Its frequent occurrence in freshwater environments and its potential toxicity towards several organisms such as fish and mussels makes DCF an emerging environmental contaminant. At typical detected environmental concentrations, the drug does not exhibit toxic effects towards living organisms, albeit chronic exposure may lead to severe effects. For DCF, about 30-70% removal has been obtained through the conventional treatment system in wastewater treatment plant being the major primary sink. Thus, the untreated DCF will pass to surface water. DCF can interact with other inorganic contaminants in the environment particularly in wastewater treatment plant, such as metals, organic contaminants and even with DCF metabolites. This process may lead to the creation of another possible emerging contaminant. In the present context, environmental fate of DCF in different compartments such as soil and water has been addressed with an overview of current treatment methods. In addition, the toxicity concerns regarding DCF in aquatic as well as terrestrial environment along with an introduction to the metabolites of DCF through consumption as well as abiotic degradation routes are also discussed. Further studies are required to better assess the fate and toxicological effects of DCF and its metabolites and must consider the possible interaction of DCF with other contaminants to develop an effective treatment method for DCF and its traces.
