      Serum microRNA-17 functions as a prognostic biomarker in osteosarcoma

          Abstract

          MicroRNAs (miRNAs/miRs) are a class of small noncoding RNA molecules that have important roles in regulating the expression of target genes associated with the development and progression of cancer. The majority of miRNAs are expressed in a highly tissue- and region-specific manner, and released into the bloodstream as a consequences of different diseases. Furthermore, altered levels of miRNAs have been observed in several diseases, including cancer. In the present study, reverse transcription-quantitative polymerase chain reaction (RT-qPCR) demonstrated that circulating miR-17 levels were significantly upregulated in patients with osteosarcoma (OS) compared with healthy subjects. RT-qPCR also revealed that high levels of circulating miR-17 expression were inversely correlated with phosphatase and tensin homolog expression, which was identified as a target gene of miR-17 in OS tissues. Furthermore, the overall survival of patients with OS was shorter in those with high miR-17 expression compared with moderate and low expression. Taken together, these findings indicate that miR-17 may function as a useful diagnostic and prognosis biomarker or therapeutic target of OS.

          PTEN, a putative protein tyrosine phosphatase gene mutated in human brain, breast, and prostate cancer.

          Mapping of homozygous deletions on human chromosome 10q23 has led to the isolation of a candidate tumor suppressor gene, PTEN, that appears to be mutated at considerable frequency in human cancers. In preliminary screens, mutations of PTEN were detected in 31% (13/42) of glioblastoma cell lines and xenografts, 100% (4/4) of prostate cancer cell lines, 6% (4/65) of breast cancer cell lines and xenografts, and 17% (3/18) of primary glioblastomas. The predicted PTEN product has a protein tyrosine phosphatase domain and extensive homology to tensin, a protein that interacts with actin filaments at focal adhesions. These homologies suggest that PTEN may suppress tumor cell growth by antagonizing protein tyrosine kinases and may regulate tumor cell invasion and metastasis through interactions at focal adhesions.

            Ribo-gnome: the big world of small RNAs.

            Small RNA guides--microRNAs, small interfering RNAs, and repeat-associated small interfering RNAs, 21 to 30 nucleotides in length--shape diverse cellular pathways, from chromosome architecture to stem cell maintenance. Fifteen years after the discovery of RNA silencing, we are only just beginning to understand the depth and complexity of how these RNAs regulate gene expression and to consider their role in shaping the evolutionary history of higher eukaryotes.

              Biology and therapeutic advances for pediatric osteosarcoma.

              Osteosarcoma is the most common malignant bone tumor in children and adolescents. Survival for these patients was poor with the use of surgery and/or radiotherapy. The introduction of multi-agent chemotherapy dramatically improved the outcome for these patients and the majority of modern series report 3-year disease-free survival of 60%-70%. This paper describes current strategies for treating patients with osteosarcoma as well as review of the clinical features, radiologic and diagnostic work-up, and pathology. The authors review the state of the art management for patients with osteosarcoma in North America and Europe including the use of limb-salvage procedures and reconstruction as well as discuss the etiologic and biologic factors associated with tumor development. Therapy-related sequelae and future directions in the biology and therapy for these patients are also discussed. Copyright AlphaMed Press
