      Antinociceptive and anti-inflammatory activities of ethanolic extract of Alafia barteri

          Abstract

          This study analyzes the antinociceptive and anti-inflammatory properties of ethanolic leaf extract of Alafia barteri Oliv., Apocynaceae, based on its medicinal use in the treatment of toothaches, inflammation and fevers. The antinociceptive effect was assessed in mice using acetic acid-induced writhing, tail clip, tail immersion and formalin assays. Anti-inflammatory activity was evaluated on carrageenan-induced paw oedema in rats, and xylene-induced ear oedema in mice. In acetic acid-induced writhing test, the extract at different doses (50, 100 and 200 mg/kg, p.o.) significantly (p < 0.05) and dose-dependently reduced pain by 35.04, 56.49 and 84.25%, respectively. The extract also significantly inhibited both the early and late phases of formalin-induced nociception in mice. In the tail immersion test, the extract caused a significant inhibition of pain (34.43% inhibition, after 90 min) at a dose of 200 mg/kg, while the effect of the extract in the tail clip test was only significant at the 100 mg/kg dose. A. barteri caused a significant inhibition of paw oedema development in the carrageenan and xylene-induced oedema tests. There was no mortality recorded following treatment with the extract (5 g/kg, p.o.). The results support the traditional use of A. barteri in the treatment of various diseases associated with pain and inflammation.

          The formalin test in mice: dissociation between inflammatory and non-inflammatory pain.

          The formalin test in mice is a valid and reliable model of nociception and is sensitive for various classes of analgesic drugs. The noxious stimulus is an injection of dilute formalin (1% in saline) under the skin of the dorsal surface of the right hindpaw. The response is the amount of time the animals spend licking the injected paw. Two distinct periods of high licking activity can be identified, an early phase lasting the first 5 min and a late phase lasting from 20 to 30 min after the injection of formalin. In order to elucidate the involvement of inflammatory processes in the two phases, we tested different classes of drugs in the two phases independently. Morphine, codeine, nefopam, and orphenadrine, as examples of centrally acting analgesics, were antinociceptive in both phases. In contrast, the non-steroid anti-inflammatory drugs indomethacin and naproxen and the steroids dexamethasone and hydrocortisone inhibited only the late phase, while acetylsalicylic acid (ASA) and paracetamol were antinociceptive in both phases. The results demonstrate that the two phases in the formalin test may have different nociceptive mechanisms. It is suggested that the early phase is due to a direct effect on nociceptors and that prostaglandins do not play an important role during this phase. The late phase seems to be an inflammatory response with inflammatory pain that can be inhibited by anti-inflammatory drugs. ASA and paracetamol seem to have actions independent of their inhibition of prostaglandin synthesis and they also have effects on non-inflammatory pain.

            Evaluation to the antioxidant activity of total flavonoids extract from persimmon (Diospyros kaki L.) leaves.

            Persimmon leaves are commonly consumed as beverages, but are also used as a popular folk medicine in China. The purpose of this work is to assess the antioxidant activity of an extract of total flavonoids from persimmon leaves (TFPL). The effect of TFPL on total antioxidant activity, reducing power, 1,1-diphenyl-2-picrylhydrazyl (DPPH()) radical scavenging, superoxide anion (()O(2)(-)) radical scavenging, hydroxyl (OH()) radical scavenging and metal chelating activities was examined. We found that TFPL possesses considerable amounts of flavonoids (192μg catechin equivalent/g of extract). The effect of this extract in total antioxidant activity, scavenging activity of superoxide anion and hydroxyl radical, reducing power and iron chelating activity was significantly better than that of rutin. However, the effect of TFPL in free radical scavenging of DPPH() was significantly not as good as than rutin. In addition, TFPL significantly decreased the level of reactive oxygen species (ROS) and malondialdehyde (MDA), while increasing the activity of catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in MC3T3-E1 cells in a dose-dependent manner. In conclusion, TFPL possess potent antioxidant and free radical scavenging activities. These antioxidant activities could contribute, at least in part, to the traditionally claimed therapeutic benefits of persimmon leaves. Copyright © 2011 Elsevier Ltd. All rights reserved.

              Analgesic and anti-inflammatory activities of [6]-gingerol.

              In the present study, the analgesic and anti-inflammatory effects of [6]-gingerol, which is the pungent constituent of ginger, were performed. Intraperitoneal administration of [6]-gingerol (25 mg/kg-50 mg/kg) produced an inhibition of acetic acid-induced writhing response and formalin-induced licking time in the late phase. [6]-Gingerol (50 mg/kg-100 mg/kg) also produced an inhibition of paw edema induced by carrageenin. These results suggested that [6]-gingerol possessed analgesic and anti-inflammatory activities.
