      Synergisms between microbial pathogens in plant disease complexes: a growing trend

          Abstract

          Plant diseases are often thought to be caused by one species or even by a specific strain. Microbes in nature, however, mostly occur as part of complex communities and this has been noted since the time of van Leeuwenhoek. Interestingly, most laboratory studies focus on single microbial strains grown in pure culture; we were therefore unaware of possible interspecies and/or inter-kingdom interactions of pathogenic microbes in the wild. In human and animal infections, it is now being recognized that many diseases are the result of multispecies synergistic interactions. This increases the complexity of the disease and has to be taken into consideration in the development of more effective control measures. On the other hand, there are only a few reports of synergistic pathogen–pathogen interactions in plant diseases and the mechanisms of interactions are currently unknown. Here we review some of these reports of synergism between different plant pathogens and their possible implications in crop health. Finally, we briefly highlight the recent technological advances in diagnostics as these are beginning to provide important insights into the microbial communities associated with complex plant diseases. These examples of synergistic interactions of plant pathogens that lead to disease complexes might prove to be more common than expected and understanding the underlying mechanisms might have important implications in plant disease epidemiology and management.

          Recent findings on the viable but nonculturable state in pathogenic bacteria.

          Many bacteria, including a variety of important human pathogens, are known to respond to various environmental stresses by entry into a novel physiological state, where the cells remain viable, but are no longer culturable on standard laboratory media. On resuscitation from this 'viable but nonculturable' (VBNC) state, the cells regain culturability and the renewed ability to cause infection. It is likely that the VBNC state is a survival strategy, although several interesting alternative explanations have been suggested. This review describes the VBNC state, the various chemical and physical factors known to induce cells into this state, the cellular traits and gene expression exhibited by VBNC cells, their antibiotic resistance, retention of virulence and ability to attach and persist in the environment, and factors that have been found to allow resuscitation of VBNC cells. Along with simple reversal of the inducing stresses, a variety of interesting chemical and biological factors have been shown to allow resuscitation, including extracellular resuscitation-promoting proteins, a novel quorum-sensing system (AI-3) and interactions with amoeba. Finally, the central role of catalase in the VBNC response of some bacteria, including its genetic regulation, is described.

            Complete viral genome sequence and discovery of novel viruses by deep sequencing of small RNAs: a generic method for diagnosis, discovery and sequencing of viruses.

            We report the first identification of novel viruses, and sequence of an entire viral genome, by a single step of high-throughput parallel sequencing of small RNAs from diseased, as well as symptomless plants. Contigs were assembled from sequenced total siRNA from plants using small sequence assembly software and could positively identify RNA, ssDNA and dsDNA reverse transcribing viruses and in one case spanned the entire genome. The results present a novel approach which cannot only identify known viral pathogens, occurring at extremely low titers, but also novel viruses, without the necessity of any prior knowledge.

              A Unified Effort to Fight an Enemy of Wheat and Barley: Fusarium Head Blight

              Wheat and barley are critical food and feed crops around the world. Wheat is grown on more land area worldwide than any other crop. In the United States, production of wheat and barley contributes to domestic food and feed use, and contributes to the export market and balance of trade. Fifteen years ago, Plant Disease published a feature article titled "Scab of wheat and barley: A re-emerging disease of devastating impact". That article described the series of severe Fusarium head blight (FHB) epidemics that occurred in the United States and Canada, primarily from 1991 through 1996, with emphasis on the unparalleled economic and sociological impacts caused by the 1993 FHB epidemic in spring grains in the Northern Great Plains region. Earlier publications had dealt with the scope and damage caused by this disease in the United States, Canada, Europe, and China. Reviews published after 1997 further described this disease and its impact on North American grain production in the 1990s. This article reviews the disease and documents the information on U.S. FHB epidemics since 1997. The primary goal of this article is to summarize a sustained, coordinated, and collaborative research program that was put in place shortly after the 1993 epidemic, a program intended to quickly lead to improved management strategies and outreach implementation. This program serves as a model to deal with other emerging plant disease threats.
