      Direct electrochemical analysis of dexamethasone endocrine disruptor in raw natural waters

          Abstract

          This paper describes an electroanalytical methodology using square-wave adsorptive voltammetry, which has been successfully applied for the direct determination of dexamethasone residues in raw natural waters used for the public supply of the Ceará State, Brazil. The obtained detection limits ranged from 7.47 × 10-9 to 1.80 × 10-8 mol L-1 for the three matrices of raw natural waters evaluated. High percentages of average recovery (98.86% ± 0.72), repeatability (0.32% ± 0.05) and reproducibility (0.91% ± 0.20) were obtained in these samples, reaffirming the sensitivity of the procedure. Energy of the LUMO orbitals and Mülliken’s atomic charges were calculated using the functional BLYP/DNP. The theoretical results allied to the diagnostic criteria of the square-wave voltammetry indicate that the dexamethasone redox mechanism is associated to the quasi-reversible and irreversible reduction process of the ketone groups located at C-20 and C-3, respectively.

          Este trabalho descreve uma metodologia eletroanalítica, utilizando voltametria adsortiva de onda quadrada, que foi aplicada com êxito na determinação direta de resíduos de dexametasona em águas naturais brutas utilizadas no abastecimento público do Estado do Ceará, Brasil. Os limites de detecção obtidos variaram entre 7,47 × 10-9 e 1,80 × 10-8 mol L-1 para as três matrizes de águas naturais brutas avaliadas. Os valores percentuais médios de recuperação (98,86% ± 0,72), repetibilidade (0,32% ± 0,05) e reprodutibilidade (0,91% ± 0,20) foram significativos, reafirmando a sensibilidade do procedimento. A energia dos orbitais LUMO e as cargas atômicas de Mülliken foram calculadas usando o funcional BLYP/DNP. Os resultados teóricos, aliados aos critérios de diagnóstico da voltametria de onda quadrada, indicam que o mecanismo redox da dexametasona está associado a processos de redução quase-reversível e irreversível dos grupos cetona localizados em C-20 e C-3, respectivamente.

            Maintenance of meiotic arrest and the induction of oocyte maturation in mouse oocyte-granulosa cell complexes developed in vitro from preantral follicles.

            During the development of oocyte-granulosa cell complexes from preantral follicles in vitro, oocytes grow and acquire competence to undergo germinal vesicle breakdown (GVB). In the culture system used here, GVB-competent oocytes were maintained in meiotic arrest solely by endogenous physiological mechanisms of the granulosa cells without supplementation with meiosis-arresting substances. Addition of mycophenolic acid, an inhibitor of inosine monophosphate (IMP) dehydrogenase, induced GVB in about 70% of the GVB-competent oocytes grown in vitro. The mechanism for meiotic arrest in this system is, therefore, similar to that for arrest in vivo insofar as it requires the participation of the IMP dehydrogenase pathway. Rp-cyclic adenosine monophosphothioate, a membrane-permeable antagonist to cAMP, induced GVB by about 30% of the competent oocytes. Cyclic AMP-dependent pathways, therefore, participate in the physiological mechanism by which mouse granulosa cells maintain meiotic arrest. Complexes were grown for 10 days in medium containing 0, 1, 5, or 10 ng/ml FSH, were stimulated with either 1 microgram/ml FSH or LH, and were assessed for GVB and cumulus expansion. GVB was stimulated by FSH whether or not the complexes were grown in medium containing FSH, but LH or hCG induced GVB only when the complexes were grown in medium containing FSH. Cumulus expansion occurred in response to either FSH or LH only when complexes were grown in medium containing FSH. FSH, therefore, promotes the differentiation of granulosa cells from preantral follicles in vitro so that LH can stimulate GVB and cumulus expansion.

              Improving sperm cryopreservation with antifreeze proteins: effect on gilthead seabream (Sparus aurata) plasma membrane lipids.

              Changes in the plasma membrane lipid composition have been related to a decrease in sperm quality during cryopreservation. Antifreeze proteins (AFPs) have been tested in different species because of their ability to depress the freezing point and their potential interaction with membranes, but controversial effects were reported. In the present study we analyzed separately the lipid composition of two sperm membrane domains, head plasma membrane (HM) and flagellar membrane (FM), after cryopreservation with an extender containing 5% dimethyl sulfoxide (DMSO) either alone or with AFPI or AFPIII (1 μg/ml). We used sperm from a teleost, Sparus aurata, because the lack of acrosome avoids changes of lipid profiles due to capacitation process or acrosomal losses during freezing/thawing. Comparing with the control (cryopreservation with 5% DMSO alone), the addition of AFPIII increased the velocity, linearity of movement, and percentage of viable cells. In addition, freezing with DMSO alone increased the phosphatidyl-serine content as well as the saturated fatty acids and decreased the unsaturated ones (mainly polyunsaturated) both in HM and FM. These changes in the lipid components were highly avoided with the addition of AFPIII. HM had a higher amount of saturated fatty acids than FM and was more affected by cryopreservation without AFPs. The percentage of viable cells was positively correlated with the amount of unsaturated fatty acids in the HM, whereas the motility parameters were positively correlated with both FM and HM amount of unsaturated fatty acids. AFPs, especially AFPIII, seem to have interacted with unsaturated fatty acids, stabilizing the plasma membrane organization during cryopreservation and contributing to improve sperm quality after thawing.
