      Premna odorata: Seasonal Metabolic Variation in the Essential Oil Composition of Its Leaf and Verification of Its Anti-Ageing Potential via In Vitro Assays and Molecular Modelling

          Abstract

          The metabolic variation in the essential oil composition of Premna odorata leaves obtained from different seasons was quantitatively and qualitatively determined employing GC/MS (Gas Chromatography coupled with Mass Spectrometry) and GC/FID (Gas chromatography equipped with flame ionization detector) techniques. It displayed the existence of 97 constituents accounting for 94.19%, 92.27%, 91.95% and 92.63% for POS (spring), POM (summer), POA (autumn) and POW (winter) whole essential oils. β-Caryophyllene constituting the main metabolite in the oil in the different seasons. To better visualize the differences between them, GC data were exposed to chemometric analysis. A PCA (principal component analysis) score plot revealed the closeness of POS and POW. Molecular modelling on collagenase, elastase and hyaluronidase enzymes active centres shows that different compounds existing in the essential oil of Premna odorata leaves shows binding to the active sites with variable degrees that suggested its anti-ageing potential. Palmitic acid displayed the highest fitting for both the collagenase and elastase active centres in both pH-based and rule-based ionization methods with ∆G equals −78.27 and −44.77 kcal/mol, respectively; meanwhile, heptacosane showed the highest fitting score in the hyaluronidase centre with ∆G = −43.78 kcal/mol. In vitro assays consolidates the obtained modelling studies in which essential oil shows considerable anti-elastase and anti-hyaluronidase potential as evidenced by their IC 50 values being 49.3 and 37.7 μg/mL, respectively; meanwhile, the essential oil of Premna odorata leaves displayed mild anti-collagenase potential. Thus, it can be concluded that Premna odorata could serve as a promising anti-ageing naturally occurring drug that could be effectively incorporated by pharmaceutical industries in cosmetics combating ageing and skin wrinkling.

          Anti-collagenase, anti-elastase and anti-oxidant activities of extracts from 21 plants

          Background Owing to their roles in tissue remodelling in health and disease, several studies have reported investigations on plant extracts as inhibitors of proteinases and as anti-oxidants. Methods The anti-ageing and anti-oxidant properties of 23 plant extracts (from 21 plant species) were assessed as anti-elastase and anti-collagenase activities and in selected anti-oxidant assays along with phenolic content. Results Anti-elastase activities were observed for nine of the extracts with inhibitory activity in the following order: white tea (~89%), cleavers (~58%), burdock root (~51%), bladderwrack (~50%), anise and angelica (~32%). Anti-collagenase activities were exhibited by sixteen plants of which the highest activity was seen in white tea (~87%), green tea (~47%), rose tincture (~41%), and lavender (~31%). Nine plant extracts had activities against both elastase (E) and collagenase (C) and were ranked in the order of white tea (E:89%, C:87%) > bladderwrack (E:50%, C:25%) > cleavers (E:58%, C:7%) > rose tincture (E:22%, C:41%) > green tea (E:10%: C:47%) > rose aqueous (E: 24%, C:26%) > angelica (E:32%, C:17%) > anise (E:32%, C:6%) > pomegranate (E:15%, C:11%). Total phenolic content varied between 0.05 and 0.26 mg gallic acid equivalents (GAE)/mL with the exception of white tea (0.77 mg GAE/mL). For anti-oxidant assessment, the Trolox equivalent anti-oxidant capacity (TEAC) assay revealed activity for all extracts. White tea had the highest activity equivalent to ~21 μM Trolox for a 6.25 μg aliquot. In addition, seven extracts exhibited activities = 10 μM Trolox with witch hazel (6.25 μg = 13 μM Trolox) and rose aqueous (6.25 μg = 10 μM Trolox) showing very high activities at low concentrations. A high activity for white tea was also found in the superoxide dismutase (SOD) assay in which it exhibited ~88% inhibition of reduction of nitroblue tetrazolium. High activities were also observed for green tea (86.41%), rose tincture (82.77%), witch hazel (82.05%) and rose aqueous (73.86%). Conclusion From a panel of twenty three plant extracts, some one dozen exhibit high or satisfactory anti-collagenase or anti-elastase activities, with nine having inhibitory activity against both enzymes. These included white tea which was found to have very high phenolic content, along with high TEAC and SOD activities.

            Plants used in cosmetics.

            This review describes the use of some natural products in cosmetic preparations, due to their low mammalian toxicity, with a brief description of the major use, plant parts used, the actives responsible for effect and the benefits of such products. Their use in skin care; such as dryness, eczema, acne, free-radical scavenging, antiinflammatory, antiaging and skin protection effects are explained, and also the use in hair care as hair growth stimulants, hair colorants, and for hair and scalp complaints such as dandruff. Essential oils when incorporated into finished products impart many benefits such as a pleasant aroma in perfumery, shine or conditioning effects in hair care products, emolliency and improving the elasticity of the skin.               In vitro determination of the anti-aging potential of four southern African medicinal plants

              Background Aging is an inevitable process for all living organisms. During this process reactive oxygen species generation is increased which leads to the activation of hyaluronidase, collagenase and elastase, which can further contribute to skin aging. Four southern African medicinal plants; Clerodendrum glabrum, Schotia brachypetala, Psychotria capensis and Peltophorum africanum, were investigated to assess their anti-aging properties. Methods Anti-elastase, anti-collagenase and anti-hyaluronidase activities of twenty-eight samples, consisting of methanol and ethyl acetate extracts of the four plants, were determined using spectrophotometric methods. Radical scavenging activity was determined by the ability of the plant extracts to scavenge the ABTS•+ radical. Results The majority of the samples in the anti-elastase assay and nine in the anti-collagenase assay showed more than 80% inhibition. The ethyl acetate extract of S. brachypetala bark and leaves of P. capensis inhibited elastase activity by more than 90%. The methanol extract of S. brachypetala bark contained the highest anti-hyaluronidase activity (75.13 ± 7.49%) whilst the ethyl acetate extract of P. africanum bark exhibited the highest antioxidant activity (IC50: 1.99 ± 0.23 μg/ml). Conclusion The free radical scavenging activity and enzyme inhibitory activity of the plant extracts investigated suggests that they can help restore skin elasticity and thereby slow the wrinkling process. P. africanum was the plant with the most promising activity and will be subjected to further testing and isolation of the active compound/s.
