      Preliminary application of DNA barcoding on rodents species investigation in mountainous regions of Datian, Fujian

          Abstract

          Objective To explore the feasibility of DNA barcoding applicate on rodents species investigation in mountainous regions of Datian, Fujian.

          Methods A total of 24 wild rodents were captured in this investigation. Morphologic identification showed that there were 14 Berylmys bowersi, 6 Bandicota indica, 3 Leopoldamys edwardsi and 1 Rattus nitidus. By DNA barcoding technology identification, there were 13 Berylmys bowersi, 6 Bandicota indica, 4 Leopoldamys edwardsi and 1 Rattus nitidus. Rodents were captured by the night trapping method. After morphological identification, liver tissue samples were collected to extract nucleic acids. The COI gene was amplified and sequenced for bioinformatics analysis. Rodents were identificated by homology comparision, genetic distance analysis and phylogenetic analysis.

          Results A total of 24 wild rodents were captured in this investigation. Morphologic identification showed that there were 14 Berylmys bowersi, 6 Bandicota indica, 3 Leopoldamys edwardsi and 1 Rattus nitidus. By DNA barcoding technology identification, there were 13 Berylmys bowersi, 6 Bandicota indica, 4 Leopoldamys edwardsi and 1 Rattus nitidus. The intraspecific genetic distance range from 0.00% to 0.21% and the interspecific genetic distance range from 13.24% to 18.03% . The interspecific genetic distance was signficantly greater than the intraspecific genetic distance.The results of DNA barcoding were differ from morphological identification in two specimens.The results of morphological identification were corrected by re-identification.

          Conclusion DNA barcoding can identify the species of rodent effectively. To explore the feasibility of DNA barcoding applicate on rodents species investigation in mountainous regions of Datian.

          Abstract

          摘要: 目的 探讨和应用基于COI的DNA条形码技术对福建省大田县野外生境进行鼠种调查。 方法 采用笼夜 法在大田县境内捕鼠, 对捕获鼠进行形态学鉴定后, 经肝脏组织核酸提取、目标条带 COI 基因片段的扩增及测序获取序 列信息, 通过同源性比对、遗传距离分析和系统进化树构建对所捕获的野栖鼠进行准确鉴定。 结果 本次调查共捕获 24只野栖鼠, 经形态学鉴定青毛鼠14只、板齿鼠6只、白腹巨鼠3只、黄毛鼠1只。经DNA条形码技术鉴定, 其中青毛鼠 13 只、板齿鼠6 只、白腹巨鼠4 只、大足鼠1 只。各鼠种种内遗传距离为0.00%~0.21%, 种间遗传距离为13.24%~ 18.03%, 种间遗传距离显著高于种内遗传距离。DNA条形码技术鉴定结果与形态学鉴定结果存在2份标本的差异, 经 再次鉴定, 更正了形态学鉴定结果。 结论 DNA条形码技术可作为形态学鉴定方法的补充, 应用于鼠种调查和分析 中。探讨和应用基于COI的DNA条形码技术对大田县山地生境进行鼠种调查。
