      Application of 9% hydrogen peroxide in final disinfection of biosafety tertiary laboratory

          Abstract

          Objective To observe the sterilization effect of 9% hydrogen peroxide and corresponding hydrogen peroxide generator on the space surface of the third-class laboratory of biosafety, and to explore the optimal sterilization scheme for insitu sterilization of the biosafety cabinet and the room exhaust high-efficiency filter.

          Methods At the same time, Bacillus subtilis and Bacillus thermophilus were used as biological indicators (1×10 6 CFU/tablet), and 9% hydrogen peroxide was used to verify the disinfection effect of the space surface and the key protective equipment (biosafety cabinet and high efficiency filter) of the biosafety level III laboratory at different times (1.5, 3.5, 5 h), so as to determine the most effective disinfection method.

          Results Keeping room temperature at 20-25 °C and relative humidity less than 55%, using 9% hydrogen peroxide and 1.5 h disinfection method can completely kill the Bacillus subtilis and thermophilic fat spore carriers on the surface of room space, but it cannot completely kill the Bacillus subtilis and thermophilic fat spore carriers on the high-efficiency filter of biological safety cabinet and the exhaust high-efficiency filter of room; using 9% hydrogen peroxide, the 3.5 h and 5 h disinfection methods can completely kill the Bacillus carrier on the surface of room space, the biological safety cabinet and the exhaust high-efficiency filter of the room.

          Conclusion 9% hydrogen peroxide and clean cell hydrogen peroxide generator can be used in the final disinfection of biosafety level III laboratory. After several disinfection tests, the best condition is: 9% hydrogen peroxide, 3.5 h.

          Abstract

          摘要: 目的 观察9%过氧化氢及相应的过氧化氢发生器对生物安全三级实验室的空间表面消毒灭菌效果, 同时探讨生物安全柜, 房间排风高效过滤器原位消毒的最佳灭菌方案。 方法 同时采用枯草芽孢杆菌和嗜热脂肪芽孢杆菌作为生物指示剂 (1×106 CFU/片), 使用9%浓度的过氧化氢对生物安全三级实验室的空间表面和实验室的关键防护设备 (生物安全柜和高效过滤器) 进行不同时间 (1.5、3.5、5 h) 的消毒效果验证, 确定最有效的消毒方法。 结果 保持室温20~25 ℃, 相对湿度小于55%, 使用9%过氧化氢, 1.5 h消毒方法可以将房间空间表面的枯草芽孢杆菌和嗜热脂肪芽孢载体完全杀灭, 但是无法将生物安全柜高效过滤器和房间的排风高效过滤器的枯草芽孢杆菌和嗜热脂肪芽孢载体完全杀灭; 使用9%过氧化氢, 3.5、5 h消毒方法可以将房间空间表面, 生物安全柜和房间的排风高效过滤器的芽孢杆菌载体完全杀灭。 结论 9%过氧化氢和配套的CLEANCube过氧化氢发生器可用于生物安全三级实验室的终末消毒, 经过多次消毒验证, 最佳条件为:9%过氧化氢, 3.5 h。
