          Abstract

          An attempt was made to evaluate microbiological quality of horticultural crops grown organically. Three species of vegetables were used, lettuce (Lactuva sativa), radish (Raphanus sativus) and spinach (Tetragonia expansa), grown organically, in fertile soil. Six different treatments were applied: mineral fertilizer, chicken, cow, and pig manure, chicken litter and cow manure, in association with a liquid foliar biofertilizer. These crops were handled according to correct agronomic practices for growing crops organically for commercial purposes. Samples were examined for the Most Probable Number (MPN/g/mL) of total and fecal coliforms and to detect the presence of Escherichia coli and Salmonella spp. All analyzed samples were considered acceptable for consumption, as Salmonella spp. was not detected. However, 63.3%, 50.0%, and 23.3% of the samples of lettuce, radish and spinach, respectively, contained >10² total coliforms/g of product. None of the samples of spinach or radish presented >10² fecal coliforms/g, and only 6.6% of lettuce samples contained >10² fecal coliforms/g. The presence of E. coli was confirmed in one sample of spinach, cultivated with cow manure.

          O objetivo do presente trabalho foi avaliar a qualidade microbiológica de hortaliças orgânicas produzidas sob diferentes condições. Três espécies de vegetais, alface (Lactuva sativa), rabanete (Raphanus sativus) e espinafre (Tetragonia expansa), foram cultivadas no sistema orgânico, em solo fertilizado com seis tratamentos diferentes: adubo mineral, estercos de galinha, bovino e suíno, cama de frango e esterco bovino associado com biofertilizante líquido de aplicação foliar. O cultivo das hortaliças foi feito de acordo com as práticas agronômicas recomendadas para o sistema orgânico em escala comercial. Das hortaliças cultivadas, foram coletadas amostras para a determinação do Número Mais Provável de coliformes totais e termotolerantes e detecção da presença de Escherichia coli e Salmonella spp. Todas as amostras analisadas foram consideradas apropriadas para o consumo humano, de acordo com a legislação brasileira em vigor, uma vez que, em nenhuma delas foi detectada a presença de Salmonella spp. Entretanto, 63,3%, 50,0% e 23,3%, respectivamente, das amostras de alface, rabanete e espinafre apresentaram contagens de coliformes totais >100/g. Em nenhuma das amostras de rabanete e espinafre o nível de coliformes termotolerantes foi >100/g e em somente 6,6% das amostras de alface foi maior que 100/g. E. coli foi detectada em uma amostra de espinafre produzido em solo fertilizado com esterco bovino.

          Pathogenic Microorganisms Associated with Fresh Produce

            Colonization of Arabidopsis thaliana with Salmonella enterica and enterohemorrhagic Escherichia coli O157:H7 and competition by Enterobacter asburiae.

            Enteric pathogens, such as Salmonella enterica and Escherichia coli O157:H7, have been shown to contaminate fresh produce. Under appropriate conditions, these bacteria will grow on and invade the plant tissue. We have developed Arabidopsis thaliana (thale cress) as a model system with the intention of studying plant responses to human pathogens. Under sterile conditions and at 100% humidity, S. enterica serovar Newport and E. coli O157:H7 grew to 10(9) CFU g(-1) on A. thaliana roots and to 2 x 10(7) CFU g(-1) on shoots. Furthermore, root inoculation led to contamination of the entire plant, indicating that the pathogens are capable of moving on or within the plant in the absence of competition. Inoculation with green fluorescent protein-labeled S. enterica and E. coli O157:H7 showed invasion of the roots at lateral root junctions. Movement was eliminated and invasion decreased when nonmotile mutants of S. enterica were used. Survival of S. enterica serovar Newport and E. coli O157:H7 on soil-grown plants declined as the plants matured, but both pathogens were detectable for at least 21 days. Survival of the pathogen was reduced in unautoclaved soil and amended soil, suggesting competition from indigenous epiphytes from the soil. Enterobacter asburiae was isolated from soil-grown A. thaliana and shown to be effective at suppressing epiphytic growth of both pathogens under gnotobiotic conditions. Seed and chaff harvested from contaminated plants were occasionally contaminated. The rate of recovery of S. enterica and E. coli O157:H7 from seed varied from undetectable to 19% of the seed pools tested, depending on the method of inoculation. Seed contamination by these pathogens was undetectable in the presence of the competitor, Enterobacter asburiae. Sampling of 74 pools of chaff indicated a strong correlation between contamination of the chaff and seed (P = 0.025). This suggested that contamination of the seed occurred directly from contaminated chaff or by invasion of the flower or silique. However, contaminated seeds were not sanitized by extensive washing and chlorine treatment, indicating that some of the bacteria reside in a protected niche on the seed surface or under the seed coat.

              Manure and Microbes: Public and Animal Health Problem?

              Most environmental concerns about waste management either have focused on the effects of nutrients, especially N and P, on water quality or have emphasized odor problems and air quality. Microbes from manure are often low on the priority list for control and remediation, despite the fact that several out-breaks of gastroenteritis have been traced to livestock operations. The pathogens discussed in this paper include protozoans (Cryptosporidium parvum, Giardia spp.), bacteria (Listeria monocytogenes, Escherichia coli O157:H7, Salmonella spp., and Mycobacterium paratuberculosis), and some enteric viruses. Clinical symptoms, prospects for zoonotic infection, and control methods other than the use of antimicrobials are considered. Recommendations to avoid disease transmission include taking steps to ensure the provision of clean, unstressful environments to reduce disease susceptibility and the careful handling and spreading of manure from animals at high risk for infection, especially young calves. Composting and drying of manure decrease the number of viable pathogens. Environmental controls, such as filter strips, also reduce the risk of water contamination.
