      Effects of Ketosis in Dairy Cows on Blood Biochemical Parameters, Milk Yield and Composition, and Digestive Capacity

          Abstract

          Introduction

          This study aimed to characterise the effects of ketosis on milk yield and composition and digestive capacity in transition dairy cows.

          Material and Methods

          Seven ketotic and seven healthy cows were housed in individual stalls for six days. Samples of plasma, milk, refused total mixed ration, and faeces were collected, and the blood biochemical parameters, milk yield and composition, dry matter intake, and faecal dry matter (FDM) production were determined.

          Results

          Compared with healthy cows, the ketotic cows had significantly higher concentrations of milk fat and citrate, but lower levels of milk protein and lactose. The cows exhibited a need for acid detergent fibre in forage and better digestion of neutral detergent fibre, starch, crude protein, and phosphorus than healthy cows, but more fat and gross energy were excreted in their faeces. Ketotic cows had higher energy-corrected milk yields and lower FDM than healthy cows.

          Conclusion

          Lower feed intake coinciding with the requirement to maintain high milk production is considered to be the cause of ketosis in dairy cows. Ketotic cows exhibited lower dry matter fat digestion.

          Prevalence of subclinical ketosis and relationships with postpartum diseases in European dairy cows.

          Subclinical ketosis (SCK) is defined as concentrations of β-hydroxybutyrate (BHBA) ≥ 1.2 to 1.4 mmol/L and it is considered a gateway condition for other metabolic and infectious disorders such as metritis, mastitis, clinical ketosis, and displaced abomasum. Reported prevalence rates range from 6.9 to 43% in the first 2 mo of lactation. However, there is a dearth of information on prevalence rates considering the diversity of European dairy farms. The objectives of this study were to (1) determine prevalence of SCK, (2) identify thresholds of BHBA, and (3) study their relationships with postpartum metritis, clinical ketosis, displaced abomasum, lameness, and mastitis in European dairy farms. From May to October 2011, a convenience sample of 528 dairy herds from Croatia, Germany, Hungary, Italy, Poland, Portugal, Serbia, Slovenia, Spain, and Turkey was studied. β-Hydroxybutyrate levels were measured in 5,884 cows with a handheld meter within 2 to 15 d in milk (DIM). On average, 11 cows were enrolled per farm and relevant information (e.g., DIM, postpartum diseases, herd size) was recorded. Using receiver operator characteristic curve analyses, blood BHBA thresholds were determined for the occurrence of metritis, mastitis, clinical ketosis, displaced abomasum, and lameness. Multivariate binary logistic regression models were built for each disease, considering cow as the experimental unit and herd as a random effect. Overall prevalence of SCK (i.e., blood BHBA ≥ 1.2 mmol/L) within 10 countries was 21.8%, ranging from 11.2 to 36.6%. Cows with SCK had 1.5, 9.5, and 5.0 times greater odds of developing metritis, clinical ketosis, and displaced abomasum, respectively. Multivariate binary logistic regression models demonstrated that cows with blood BHBA levels of ≥ 1.4, ≥ 1.1 and ≥ 1.7 mmol/L during 2 to 15 DIM had 1.7, 10.5, and 6.9 times greater odds of developing metritis, clinical ketosis, and displaced abomasum, respectively, compared with cows with lower BHBA blood levels. Interestingly, a postpartum blood BHBA threshold ≥ 1.1 mmol/L increased the odds for lameness in dairy cows 1.8 (95% confidence interval: 1.3 to 2.5) times. Overall, prevalence of SCK was high between 2 to 15 DIM and SCK increased the odds of metritis, clinical ketosis, lameness, and displaced abomasum in European dairy herds.

            The effect of subclinical ketosis in early lactation on reproductive performance of postpartum dairy cows.

            Data generated from 796 Holstein cows enrolled in a clinical trial to investigate the health effect of a monensin controlled release capsule were analyzed to investigate the association between circulating serum beta-hydroxybutyrate (BHBA) concentration in the peri-parturient period and subsequent reproductive performance. Overall, accounting for both repeated measures within cow and clustering at the herd level, non-pregnant cows after first insemination tended to have increased circulating BHBA concentrations from 3 wk before calving to 9 wk after calving relative to pregnant cows. Including the interaction between the week of sample collection and pregnancy outcome, non-pregnant cows had higher circulating BHBA concentrations in the second week after calving than cows diagnosed pregnant after first artificial insemination. Within individual weeks, cows with circulating BHBA concentrations > or =1,000 micromol/L in the first week postpartum were less likely to be diagnosed pregnant after first insemination. In the second week postpartum, the cows with circulating BHBA concentrations > or =1,400 micromol/L were significantly less likely to be pregnant after first artificial insemination. A dose response relationship was found when a comparison of the probability of pregnancy after first insemination and duration of elevated circulating ketone bodies was investigated. The probability of pregnancy was reduced by 20% in cows diagnosed subclinically ketotic in either the first or second week postpartum. Nevertheless, cows above the subclinical ketosis threshold in both the first and second week postpartum were 50% less likely to be pregnant after first insemination. Similarly, the median time to pregnancy increased in cows experiencing elevated BHBA concentrations in either (124 d) or both (130 d) the first and second week postpartum relative to cows never experiencing elevated BHBA concentrations (108 d). To further investigate this, the effect of elevated circulating BHBA was permitted to vary with time. The effect decreased with time, such that the daily probability of pregnancy increased similar to nonsubclinically ketotic cows by approximately 160 d in milk. From this analysis, both the relative circulating concentration of BHBA and the duration of elevated circulating BHBA were negatively associated with the probability of pregnancy at first service.

              Relationships among milk yield, body condition, cow weight, and reproduction in spring-calved Holstein-Friesians.

              Relationships among milk production, body condition score (BCS), body weight (BW), and reproduction were studied using logistic regression on data from 6433 spring-calving Holstein-Friesian dairy cows in 74 commercial herds. Multivariate models were adjusted for herd, breeding value for milk yield, proportion of Holstein-Friesian genes, lactation number, calving period, and degree of calving assistance. Significant associations between reproductive measures and components of energy balance were identified. Higher 200-d milk protein content and higher protein-to-fat ratio at start of breeding were associated with increased likelihood of submission for breeding in the first 21 d of the breeding season (SR21). High 100-d cumulative milk yield as a proportion of estimated 305-d milk yield (low persistency) was associated with a lower likelihood of pregnancy to first service (PREG1), whereas cows reaching peak milk yields earlier tended to have higher PREG1. Cows that reached nadir milk protein content relatively late in lactation had lower PREG1. Milk yield at first service and 305-d milk protein content were positively associated with the likelihood of pregnancy after 42 d of breeding (PR42). Higher 305-d milk lactose content was associated with increased PREG1 and PR42. Mean BCS at 60 to 100 d of lactation was positively associated with both SR21 and PR42, whereas nadir BCS was positively associated with PREG1. Cows with precalving BCS > 3.0 that also lost > 0.5 BCS unit by first service had lower PR42. More BW gain for 90 d after start of breeding was associated with higher SR21 and PREG1; more BW gain for 90 d after first service was associated with higher PR42. Milk protein and lactose content, BCS, and BW changes are important tools to identify cows at risk of poor reproduction.
