      Simultaneous Determination of Arbutin and Hydroquinone in Different Herbal Slimming Products Using Gas Chromatography-mass Spectrometry

          Abstract

          Objectives:

          The aim of our study was to develop a simple, precise, sensitive and specific method for the simultaneous determination of arbutin and hydroquinone in different herbal slimming products using GC-MS.

          Materials and Methods:

          The methanol and aqueous extracts of nine herbal slimming products in Turkey were evaluated for analysis of arbutin and hydroquinone using GC-MS method.

          Results:

          The retention times of arbutin and hydroquinone were found as 11.32 and 5.44 min, respectively. The linear ranges in this method were 5-500 ng/mL for arbutin and hydroquinone, respectively. The intra- and inter-day precisions, expressed as the relative standard deviation, were less than 1.94 and 2.73%, determined from quality control samples for arbutin and hydroquinone, and accuracy was within 1.13 and 2.56% in terms of relative error, respectively. The limit of detection and quantification were 0.555 and 1.665 ng/mL for arbutin, and 0.031 and 0.093 ng/mL for hydroquinone, respectively.

          Conclusion:

          The developed method can be used for routine quality control analysis of arbutin and hydroquinone in different herbal slimming products.

            MAO-A inhibitory activity of quercetin from Calluna vulgaris (L.) Hull.

            This study investigated MAO-A inhibitory activity of methanol extract of Calluna vulgaris (L.) Hull., which traditionally has been used as a nerve calming remedy. A methanolic extract of Calluna vulgaris was partitioned against heptane, ethyl acetate and water. The three fractions were tested in a photometric peroxidase linked MAO-A bioassay. The ethyl acetate phase showed the highest MAO-A inhibitory activity. Quercetin was isolated by VLC through bioassay-guided fractionation and purified by re-crystallisation. The structure was elucidated by LC-MS and (1)H NMR. The IC(50)-value for MAO-A inhibition by quercetin was 18+/-0.2 microM in an assay where the IC(50)-value for MAO-A inhibition by clorgylin was 0.2+/-0.02 microM. The content of quercetin in Calluna vulgaris might explain the reported nerve calming effect of the plant.

              Quantitative Determination of Arbutin and Hydroquinone in Different Plant Materials by HPLC

              A simple, fast method of high-performance liquid chromatography for the determination and quantification of arbutin and hydroquinone in many different raw materials was developed and validated. The optimum conditions for the separation and detection of these two constituents were achieved on a LiChro-CARD 125-4 Superspher®100 RP-18 column with the water-methanol (gradient elution) mobile phase and recorded at 289 nm. The purities of peaks were verified by PDA analysis of impurities. The results of validation have shown that the HPLC method is stable and accurate for the simultaneous determination of arbutin and hydroquinone in extracts from various plants. The developed method gave a good sensitivity (LOD 1µg/ml for arbutin and 0.49 µg/ml for hydroquinone) with linearity R2 >0.9993 (for both). The relative standard deviation of the method was less than 2.53% for intra-day assays and 3.23% for inter-day assay, the accuracy of the recovery test ranged from 98.96% to 106.4%. This method was used in comparative qualitative analysis of arbutin and hydroquinone in 16 different raw materials from families Lamiaceae, Ericacaeae, Saxifragaceae, Rosaceae. The content of arbutin in B. ciliata, B. cordifolia and Ledum palustre was examined for the first time.
