          Abstract

          La determinación mediante espectroscopia de infrarrojo cercano de atributos críticos de calidad, de interés en la fabricación industrial de sólidos farmacéuticos, es estudiada con el objetivo de aplicar esta nueva metodología más rápida y eficazmente como una alternativa a los métodos convencionales. Se estudia el proceso de mezclado de los componentes de una fórmula efervescente con el fin de determinar el tiempo final del mezclado de una forma rápida y confiable, así como la humedad en los granulados intermedios de tabletas masticables. El objetivo es construir la calidad del producto final, controlando atributos críticos de calidad de los productos intermedios en el mismo proceso aplicando la Tecnología Analítica de Procesos (PAT).

          The determination by means of near infrared spectroscopy of critical quality attributes, of interest for the industrial manufacture of pharmaceutical solids, is studied here in order to apply this new methodology more quickly and efficiently as an alternative to conventional methods. The mixing process of the components of an effervescent formula is studied in order to determine the final mixing time in a fast and reliable way, as well as the humidity in intermediate granules of chewable tablets. The objective is to build the quality of the final product, controlling critical quality attributes of intermediate products during the same process by applying Process Analytical Technology (PAT)

          Effect of particle properties on the flowability of ibuprofen powders.

          Powder flowability is one of the key parameters in the pharmaceutical tabletting process. The flowability is affected by both the particles' properties and the tabletting equipment characteristics. Although it is generally accepted that powder flowability increases with an increase in particle size, quantitative studies and comprehensive theoretical insights into the particle property effects are still lacking. In this paper, ibuprofen, a non-steroidal drug widely used as an anti-inflammatory analgesic was chosen as a model material to assess the effect of particle properties on its flowability. Ibuprofen typically has a needle shaped morphology. The flowability of ibuprofen size fractions was studied in detail using two flow measurement methods. The separated fractions were also compared to magnesium stearate lubricated ibuprofen and its size fractions. The experimental results showed that powder flowability is significantly affected by both the particle size and size distribution. The finest size fraction that is separated from the bulk ibuprofen powder flows better than the bulk powder. For powders with narrow size distributions, the flowability increases significantly with the increase in particle size. In addition, admixing magnesium stearate to ibuprofen not only increases the flow function of the powder, but also reduces the internal friction angle. A theoretical analysis based on the limiting tensile strength of the powder bed was carried out and the flow conditions for particles of different size and shape were developed.

            In-line moisture measurement during granulation with a four-wavelength near infrared sensor: an evaluation of particle size and binder effects.

            Factors affecting in-line near infrared (NIR) moisture measurement with a four-wavelength sensor were evaluated (choice of binder used in granulation liquid and the increase in particle size). An entire NIR spectrum is not necessary for the measurement of water, and often the use of only a few NIR wavelengths around the water band enables reliable and high-speed detection of moisture. Glass ballotini and microcrystalline cellulose (MCC) were used as model test materials. The binders studied were poly[1-(2-oxo-1-pyrrolidinyl)ethylene] (PVP) and gelatin. Full off-line NIR spectra of test materials at different levels of binder solutions were measured. The major spectral features for both the binders were bands around 1700 nm (first overtones CH related stretches) and 2200 nm (combination bands). Gelatin also had an NH band around 1500 nm (first overtones of NH stretches) and combination bands at about 2050 nm. Particle size effects were observed as an increase in spectra baseline. All these factors should be considered when choosing NIR wavelengths used for detection of water with a fixed wavelength set-up. A robust calibration model enables the development of in-process control of wet granulation processes.

              Controlling granule size by granulation liquid feed pulsing.

              The effects of inlet air humidity, granulation liquid feed rate and granulation liquid feed pulsing on the particle-size distribution of final granules were studied in a laboratory scale fluid-bed granulator using a central composite study design. The factors examined were modelled using three different particle-size measurement techniques (sieve analysis, laser light diffraction and the spatial filtering technique). Best goodness of fit (R2=0.94) and goodness of prediction (Q2=0.90) values were obtained using particle-size results of the spatial filtering technique. Increasing inlet air humidity and granulation liquid feed rate resulted in greater median granule size, as expected. When pulsing (interruption of granulation liquid feed in predetermined sequences) was used, the median granule size decreased clearly. This effect was strong, especially with high inlet air humidity and rapid liquid feed rate processes. Granulation liquid feed pulsing is an effective way to modify the particle size of final granules. Pulsing can be used as a controlling tool to compensate for excessive moisture content in the granulation process.
