          Abstract

          Se estudió la propagación in vitro de Nothofagus procera sobre los medios de cultivo MS y BTM usando embriones maduros como material inicial. En la fase de establecimiento embrionario, pese a no haber diferencias estadísticas, se optó por el medio MS, ya que en éste las microplántulas mostraron mayor vigor. En una posterior etapa de proliferación, los resultados mostraron que los tratamientos hormonales, en especial 0,5 mgl-1 de BAP y 0,05 mgl-1 de AIB son significativamente mejores para la inducción de yemas y tallos. Se observó que muchos puntos de crecimiento no elongaron a estas concentraciones, lo cual se superó al estrechar la relación citoquinina/ auxina a 0,5 mgl-1 de BAP más 0,07 mgl-1 de AIB. Sin embargo, la manifestación y crecimiento de los órganos inducidos es significativamente mejor en medio MS sin hormonas, donde se logra, incluso, rizogénesis adventicia en forma espontánea.

          Translated abstract

          In vitro propagation of Nothofagus procera was studied in MS and BTM culture media using mature embryos as the initiation material. Even at the embrionary establishment phase, there was no statistical difference between two media and the MS medium was selected because it produced more vigorous microseedlings. In the following proliferation phase, the results showed that hormonal treatments, in particular 0.5 mg/l of BAP and 0.05 mg/l of IBA, were better for the induction of buds and shoots. There were not many growth points observed at this concentration, which was improved by increasing the hormonal concentration to 0.5 mg/l of BAP and 0.07 mg/l of IBA. However, the manifestation and growth of the induced organs was significantly better in MS medium without hormones, which also spontaneously formed roots.

              Endogenous polyamine concentrations in juvenile, adult and in vitro reinvigorated hazel.

              We investigated endogenous polyamine concentrations in leaves from juvenile and mature hazel (Corylus avellana L.) shoots, as well as leaves from shoots obtained by both forced outgrowth and micropropagation of adult tissues. To determine if the observed in vitro reinvigoration was associated with polyamine metabolism, we tested the effect of serial subcultures on polyamine concentrations. Polyamines, mostly free putrescine, were higher in juvenile tissues. Adult tissues micropropagated for 14 subcultures had polyamine concentrations characteristic of juvenile tissues. However, with additional subcultures, total polyamine concentrations decreased. The putrescine to spermidine plus spermine ratio was higher in juvenile and micropropagated tissues than in adult tissues, but decreased in micropropagated tissues after 20 subcultures. This ratio may reflect a balance between vegetative growth and reproductive processes. Thus, an analysis of polyamine concentrations may provide a simple assay for determining the juvenility of plant tissues and, hence, their suitability for micropropagation.
