      Impacts of Extraction Methods in the Rapid Determination of Atrazine Residues in Foods using Supercritical Fluid Chromatography and Enzyme-Linked Immunosorbent Assay: Microwave Solvent vs. Supercritical Fluid Extractions

          Abstract

          It is an accepted fact that many food products that we eat today have the possibility of being contaminated by various chemicals used from planting to processing. These chemicals have been shown to cause illnesses for which some concerned government agencies have instituted regulatory mechanisms to minimize the risks and the effects on humans. It is for these concerns that reliable and accurate rapid determination techniques are needed to effect proper regulatory standards for the protection of people's nutritional health. This paper, therefore, reports the comparative evaluation of the extraction methods in the determination of atrazine (commonly used in agricultural as a herbicide) residues in foods using supercritical fluid chromatography (SFC) and enzyme-linked immunosorbent assay (ELISA) techniques. Supercritical fluid extraction (SFE) and microwave solvent extraction (MSE) methods were used to test samples of frozen vegetables, fruit juice, and jam from local food markets in Houston. Results showed a high recovery percentage of atrazine residues using supercritical fluid coupled with ELISA and SFC than with MSE. Comparatively, however, atrazine was detected 90.9 and 54.5% using SFC and ELISA techniques, respectively. ELISA technique was, however, less time consuming, lower in cost, and more sensitive with low detection limit of atrazine residues than SFC technique.

          Dissipation and distribution of atrazine, simazine, chlorpyrifos, and tetradifon residues in citrus orchard soil.

          An environmental fate study was conducted in a citrus orchard plot in València (Spain) in the fall of 1993. Dissipation and distribution of atrazine, simazine, chlorpyrifos and tetradifon residues following their controlled addition for agricultural purposes in a mediterranean red soil (Luvic Calcisol, Rhodoxeralf) were evaluated. During a two-month period, the amounts of applied pesticides in different soil layers (0-0.05, 0.05-0.22, 0.22-0.42, and 0.42-0.52 m) were monitored. In addition, information on soils, weather and agricultural practice were collected. Degradation half-lives were calculated, assuming zero-order kinetics: 11 days for atrazine, 12 days for simazine, 10 days for chlorpyrifos, and 18 for tetradifon. The distribution through the soil profile shows that the pesticide concentrations were always highest in the upper layer (0-0.05 m) of soil, and that atrazine was the most mobile of all the four pesticides investigated.

            Supercritical fluid extraction of atrazine and other triazine herbicides from fortified and incurred eggs.

            Triazines are a class of important pre-emergent weed herbicides. Some members of this class of herbicides exhibit carcinogenic and immunotoxicity properties, which make their use controversial in areas where animal feed crops are grown. It is therefore important to determine if triazine residues are transported to animal food products in order to ascertain the extent of human exposure. Most of the current herbicide residue extraction methods are time-consuming and solvent intensive. Supercritical fluid extraction (SFE) using CO(2) has been used as a alternative for other residue extraction methods as a replacement for hazardous organic solvents. In this study, 10 triazines were extracted from eggs fortified at 100 ppb using unmodified supercritical CO(2) at a pressure of 10000 psi and a temperature of 50 degrees C with off-line collection on a solid phase extraction cartridge containing Florisil. Atrazine recovery averaged 90.4% with an RSD of 3.3%. The other triazines were recovered at mean levels >73%. In a separate feeding study, atrazine and two of its dealkyl metabolites were detected in the egg. The results indicate that SFE is a viable technique for isolating triazine residues from eggs, requiring only 8 mL of solvent for each analysis.
