          Abstract

          An epidemiological analysis of the temporal and spatial patterns of LSD epidemics and of the risk factors for LSD spread in south‐eastern Europe was performed, based on the data collected from affected and at risk countries. Since 2015, the extent of the LSD epidemics in south‐eastern Europe was over 7,600 LSD outbreaks with 12,800 affected animals, with most outbreaks occurring between May and August. Most LSD spread occurs over a relatively small distance, approximately between 10 and 20 km, and the speed of propagation was estimated to be mostly up 2 km/day, in agreement with the vector‐borne pattern of LSD. Proximity to affected farms, warm temperatures and related vector abundance were among the main risk factors for LSD spread. Within a few months’ at least 90% of the animal population had been vaccinated with live homologous vaccine against LSD in south‐eastern Europe. Where almost total vaccination coverage was achieved, no further outbreaks were reported. The vaccination effectiveness in Albania was estimated to be around 70% at farm level and 77% at animal level. Possible adverse effects to live homologous vaccine, including fever, decreased milk production and oedema at injection site were reported in Croatia (a LSD‐free country) mostly within 2 weeks after vaccination, in 0.09% of the vaccinated animals. Unique farm identifiers should be always used across all databases, so to allow further analysis especially on improving the mathematical models for more robust estimates of transmission parameters applicable to the region, and for better estimation of vaccination effectiveness. All suspected clinical cases in vaccinated animals should be confirmed by differentiating field virus from vaccine strain. Trapping surveys for estimation of vector abundance can be carried out by targeting some sentinel farms, to be followed up during the whole LSD season, while long‐term studies can give more accurate information about species composition and seasonality of potential LSD vectors.

          Morphological and genetic diversity of Rhipicephalus sanguineus sensu lato from the New and Old Worlds

          Background The taxonomic status of the brown dog tick (Rhipicephalus sanguineus sensu stricto), which has long been regarded as the most widespread tick worldwide and a vector of many pathogens to dogs and humans, is currently under dispute. Methods We conducted a comprehensive morphological and genetic study of 278 representative specimens, which belonged to different species (i.e., Rhipicephalus bursa, R. guilhoni, R. microplus, R. muhsamae, R. pusillus, R. sanguineus sensu lato, and R. turanicus) collected from Europe, Asia, Americas, and Oceania. After detailed morphological examination, ticks were molecularly processed for the analysis of partial mitochondrial (16S rDNA, 12S rDNA, and cox1) gene sequences. Results In addition to R. sanguineus s.l. and R. turanicus, three different operational taxonomic units (namely, R. sp. I, R. sp. II, and R. sp. III) were found on dogs. These operational taxonomical units were morphologically and genetically different from R. sanguineus s.l. and R. turanicus. Ticks identified as R. sanguineus s.l., which corresponds to the so-called “tropical species” (=northern lineage), were found in all continents and genetically it represents a sister group of R. guilhoni. R. turanicus was found on a wide range of hosts in Italy and also on dogs in Greece. Conclusions The tropical species and the temperate species (=southern lineage) are paraphyletic groups. The occurrence of R. turanicus in the Mediterranean region is confirmed. A consensual re-description of R. sanguineus s.s. and R. turanicus will be necessary to solve the taxonomic problems within the so-called R. sanguineus group.

            Development and validation of IIKC: an interactive identification key for Culicoides (Diptera: Ceratopogonidae) females from the Western Palaearctic region

            Background and methods The appearance of bluetongue virus (BTV) in 2006 within northern Europe exposed a lack of expertise and resources available across this region to enable the accurate morphological identification of species of Culicoides Latreille biting midges, some of which are the major vectors of this pathogen. This work aims to organise extant Culicoides taxonomic knowledge into a database and to produce an interactive identification key for females of Culicoides in the Western Palaearctic (IIKC: Interactive identification key for Culicoides). We then validated IIKC using a trial carried out by six entomologists based in this region with variable degrees of experience in identifying Culicoides. Results The current version of the key includes 98 Culicoides species with 10 morphological variants, 61 descriptors and 837 pictures and schemes. Validation was carried out by six entomologists as a blind trial with two users allocated to three classes of expertise (beginner, intermediate and advanced). Slides were identified using a median of seven steps and seven minutes and user confidence in the identification varied from 60% for failed identifications to a maximum of 80% for successful ones. By user class, the beginner group successfully identified 44.6% of slides, the intermediate 56.8% and the advanced 74.3%. Conclusions Structured as a multi-entry key, IIKC is a powerful database for the morphological identification of female Culicoides from the Western Palaearctic region. First developed for use as an interactive identification key, it was revealed to be a powerful back-up tool for training new taxonomists and to maintain expertise level. The development of tools for arthropod involvement in pathogen transmission will allow clearer insights into the ecology and dynamics of Culicoides and in turn assist in understanding arbovirus epidemiology.

              The development of a multipurpose trap (the Nzi) for tsetse and other biting flies.

              New trap designs for tsetse (Glossinidae), stable flies (Muscidae: Stomoxyinae), and horse flies (Tabanidae) were tested in Kenya to develop a multipurpose trap for biting flies. Many configurations and colour/fabric combinations were compared to a simplified, blue-black triangular trap to identify features of design and materials that result in equitable catches. New designs were tested against conventional traps, with a focus on Glossina pallidipes Austen and G. longipennis Corti, Stomoxys niger Macquart, and Atylotus agrestis (Wiedemann). A simple design based on minimal blue and black rectangular panels, for attraction and contrast, with a trap body consisting of an innovative configuration of netting, proved best. This 'Nzi' trap (Swahili for fly) caught as many or significantly more tsetse and biting flies than any conventional trap. The Nzi trap represents a major improvement for Stomoxyinae, including the cosmopolitan species S. calcitrans (Linnaeus), with up to eight times the catch for key African Stomoxys spp. relative to the best trap for this group (the Vavoua). Catches of many genera of Tabanidae, including species almost never caught in traps (Philoliche Wiedemann), are excellent, and are similar to those of larger traps designed for this purpose (the Canopy). Improvements in capturing biting flies were achieved without compromising efficiency for the savannah tsetse species G. pallidipes. Catches of fusca tsetse (G. longipennis and G. brevipalpis Newstead) were higher or were the same as catches in good traps for these species (NG2G, Siamese). Altogether, the objective of developing a simple, economical trap with harmonized efficiency was achieved.
