      Preparation and Characterization of Novel Green Seaweed Films from Ulva rigida

          Abstract

          Ulva rigida green seaweed is an abundant biomass consisting of polysaccharides and protein mixtures and a potential bioresource for bioplastic food packaging. This research prepared and characterized novel biodegradable films from Ulva rigida extracts. The water-soluble fraction of Ulva rigida was extracted and prepared into bioplastic films. 1H nuclear magnetic resonance indicated the presence of rhamnose, glucuronic and sulfate polysaccharides, while major amino acid components determined via high-performance liquid chromatography (HPLC) were aspartic acid, glutamic acid, alanine and glycine. Seaweed extracts were formulated with glycerol and triethyl citrate (20% and 30%) and prepared into films. Ulva rigida films showed non-homogeneous microstructures, as determined via scanning electron microscopy, due to immiscible crystalline component mixtures. X-ray diffraction also indicated modified crystalline morphology due to different plasticizers, while infrared spectra suggested interaction between plasticizers and Ulva rigida polymers via hydrogen bonding. The addition of glycerol decreased the glass transition temperature of the films from −36 °C for control films to −62 °C for films with 30% glycerol, indicating better plasticization. Water vapor and oxygen permeability were retained at up to 20% plasticizer content, and further addition of plasticizers increased the water permeability up to 6.5 g·mm/m2·day·KPa, while oxygen permeability decreased below 20 mL·mm/m2·day·atm when blending plasticizers at 30%. Adding glycerol efficiently improved tensile stress and strain by up to 4- and 3-fold, respectively. Glycerol-plasticized Ulva rigida extract films were produced as novel bio-based materials that supported sustainable food packaging.

          Robust superhydrophobicity: mechanisms and strategies

          By incorporating multiple strategies, passive resistance and active regeneration, robust superhydrophobicity has been explored via various approaches for diverse applications. Superhydrophobic surfaces hold great prospects for extremely diverse applications owing to their water repellence property. The essential feature of superhydrophobicity is micro-/nano-scopic roughness to reserve a large portion of air under a liquid drop. However, the vulnerability of the delicate surface textures significantly impedes the practical applications of superhydrophobic surfaces. Robust superhydrophobicity is a must to meet the rigorous industrial requirements and standards for commercial products. In recent years, major advancements have been made in elucidating the mechanisms of wetting transitions, design strategies and fabrication techniques of superhydrophobicity. This review will first introduce the mechanisms of wetting transitions, including the thermodynamic stability of the Cassie state and its breakdown conditions. Then we highlight the development, current status and future prospects of robust superhydrophobicity, including characterization, design strategies and fabrication techniques. In particular, design strategies, which are classified into passive resistance and active regeneration for the first time, are proposed and discussed extensively.

            Immunomodulating activities of acidic sulphated polysaccharides obtained from the seaweed Ulva rigida C. Agardh.

            Water-soluble acidic polysaccharides from the cell walls of Ulva rigida are mainly composed of disaccharides that contain glucuronic acid and sulphated rhamnose. The structure of disaccharides resembles that of glycosaminoglycans (GAGs) as they both contain glucuronic acid and sulphated sugars. Glycosaminoglycans occur in the extracellular matrix of animal connective tissues but can also be produced by leucocytes at inflammatory sites. Certain types of GAGs can even activate macrophages and therefore the acidic polysaccharides from U. rigida probably modulate macrophage activity. In the present study, we evaluated the effects of U. rigida polysaccharides on several RAW264.7 murine macrophage activities, including expression of inflammatory cytokines and receptors, nitric oxide and prostaglandin E2 (PGE(2)) production, and nitric oxide synthase 2 (NOS-2) and cyclooxygenase-2 (COX-2) gene expression. U. rigida acidic polysaccharides induced a more than two-fold increase in the expression of several chemokines (chemokine (C motif) ligand 1, chemokine (C-X-C motif) ligand 12, chemokine (C-C motif) ligand 22 and chemokine (C-X-C motif) ligand 14 (Cxcl14)) and in the expression of IL6 signal transducer and IL12 receptor beta 1. Incubation of macrophages with U. rigida polysaccharides also induced an increase in nitrite production, although this effect decreased considerably after desulphation of polysaccharides, suggesting that the sulphate group is important for the stimulatory capacity of these molecules. U. rigida polysaccharides also stimulated macrophage secretion of PGE(2) and induced an increase in COX-2 and NOS-2 expression. The results indicate that U. rigida acid polysaccharide can be used as an experimental immunostimulant for analysing inflammatory responses related to macrophage functions. In addition, these polysaccharides may also be of clinical interest for modifying certain macrophage activities in diseases where macrophage function is impaired or needs to be boosted.

              “Bligh and Dyer” and Folch Methods for Solid–Liquid–Liquid Extraction of Lipids from Microorganisms. Comprehension of Solvatation Mechanisms and towards Substitution with Alternative Solvents

              Bligh and Dyer (B & D) or Folch procedures for the extraction and separation of lipids from microorganisms and biological tissues using chloroform/methanol/water have been used tens of thousands of times and are “gold standards” for the analysis of extracted lipids. Based on the Conductor-like Screening MOdel for realistic Solvatation (COSMO-RS), we select ethanol and ethyl acetate as being potentially suitable for the substitution of methanol and chloroform. We confirm this by performing solid–liquid extraction of yeast (Yarrowia lipolytica IFP29) and subsequent liquid–liquid partition—the two steps of routine extraction. For this purpose, we consider similar points in the ternary phase diagrams of water/methanol/chloroform and water/ethanol/ethyl acetate, both in the monophasic mixtures and in the liquid–liquid miscibility gap. Based on high performance thin-layer chromatography (HPTLC) to obtain the distribution of lipids classes, and gas chromatography coupled with a flame ionisation detector (GC/FID) to obtain fatty acid profiles, this greener solvents pair is found to be almost as effective as the classic methanol–chloroform couple in terms of efficiency and selectivity of lipids and non-lipid material. Moreover, using these bio-sourced solvents as an alternative system is shown to be as effective as the classical system in terms of the yield of lipids extracted from microorganism tissues, independently of their apparent hydrophilicity.
