      Evaluation of in-vitro Antioxidant Properties of Hydroalcoholic Solution Extracts Urtica dioica L., Malva neglecta Wallr. and Their Mixture

          Abstract

          The study was aimed at evaluating the antioxidant activity of hydroalcoholic solution extracts of Urtica dioica L. (UD), Malva neglecta Wallr. (MN) plants and their mixture. In this study, flower (UDF), root (UDR), seed (UDS) and leaf (UDL) parts of UD and flower (MNF) and leaf (MNL) parts of MN were used. The antioxidant properties of hydroalcoholic extracts and their mixture were evaluated using different antioxidant tests such as total antioxidant activity, reducing power, superoxide anion radical scavenging, hydrogen peroxide scavenging, free radical scavenging, and metal chelating activity for comparison. In addition, total phenolic compounds in the extracts of both plants were determined as catechin equivalent. The various antioxidant activities were compared to natural and synthetic standard antioxidants such as BHA, BHT and α-tocopherol. According to FTC method, the both extracts exhibited strong total antioxidant activity. At the concentration of 100 μg/mL, Hydroalcoholic extracts of UDS, UDR, UDF, UDL, MNF, MNL, and UD-MN showed 81.7%, 79.8%, 78.3%, 76.4%, 77.3%, 74.1%, and 80.7%, respectively. Comparable, 100 μg/mL of standard antioxidants BHA, BHT and α-tocopherol exhibited 66.2%, 70.6%, and 50.1% inhibition on peroxidation of linoleic acid emulsion, respectively. In addition, UD-MN showed strong superoxide anion radical scavenging activity comparable with UDR, UDF, UDL, MNF, and MNL. Based on the findings, plants mixture was commonly found to have synergistically higher antioxidant activity.

          Prevention of cytotoxicity and inhibition of intercellular communication by antioxidant catechins isolated from Chinese green tea.

          An antioxidant fraction of Chinese green tea (green tea antioxidant; GTA), containing several catechins, has been previously shown to inhibit 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced tumor promotion in mouse skin. In the present study, GTA was shown to have antioxidative activity toward hydrogen peroxide (H2O2) and the superoxide radical (O2-). GTA also prevented oxygen radical and H2O2-induced cytotoxicity and inhibition of intercellular communication in cultured B6C3F1 mouse hepatocytes and human keratinocytes (NHEK cells). GTA (0.05-50 micrograms/ml) prevented the killing of hepatocytes (measured by lactate dehydrogenase release) by paraquat (1-10 mM) and glucose oxidase (0.8-40 micrograms/ml) in a concentration-dependent fashion. GTA (50 micrograms/ml) also prevented the inhibition of hepatocyte intercellular communication by paraquat (5 mM), glucose oxidase (0.8 micrograms/ml), and phenobarbital (500 micrograms/ml). In addition, GTA (50 micrograms/ml) prevented the inhibition of intercellular communication in human keratinocytes by TPA (100 ng/ml). Cytotoxicity and inhibition of intercellular communication, two possible mechanisms by which tumor promoters may produce their promoting effects were therefore prevented by GTA. The inhibition of these two effects of pro-oxidant compounds may suggest a mechanism by which GTA inhibits tumor promotion in vivo.

            Free radicals as mediators of tissue injury and disease.

            A radical is any molecule that contains one or more unpaired electrons. Radicals are normally generated in many metabolic pathways. Some of these radicals can exist in a free form and subsequently interact with various tissue components resulting in dysfunction. The potential role of oxygen- or xenobiotic-derived free radicals in the pathology of several human diseases has stimulated extensive research linking the toxicity of numerous xenobiotics and disease processes to a free radical mechanism. However, because free radical-mediated changes are pervasive and often poorly understood, the question of whether such species are a major cause of tissue injury and human disease remains equivocal. This review discusses cellular sources of various radical species and their reactions with vital cellular constituents. Examples of purported free radical-mediated disorders are discussed in detail to provide insights into the controversy over whether free radicals are important mediators of tissue injury.

              Free radical scavenging activities of mushroom polysaccharide extracts.

              The superoxide and hydroxyl radical scavenging activities of eight mushroom antitumor polysaccharide extracts were investigated using phenazin methosulphate-NADH-nitroblue tetrazolium system and ascorbic acid-Cu(2+)-cytochrome C system respectively. The results showed that six of eight mushroom polysaccharide extracts had superoxide and hydroxyl radical scavenging activities. The protein content of the polysaccharide extracts appeared to contribute a direct effect on free radical scavenging activity. However, none of the mushroom polysaccharide extracts had antioxidative activity as measured by detecting malondialdehyde (MDA) contents of liver microsomes.
